Effect of Shencao Tongmai Granule(参草通脉颗粒)on Ventricular Remodeling in Rats with Chronic Heart Failure

摘  要:目的探讨参草通脉颗粒对慢性心力衰竭(慢性心衰)大鼠MMP-2、TIMP-2、AngⅡ、ACE2及心室重构的影响。方法80只大鼠从中随机选取15只大鼠作为假手术组,65只构建慢性心衰模型,成模后随机选取44只,模型组8只,西药组及中药高、中、低剂量组,每组各9只;采用蒸馏水灌胃模型组、假手术组大鼠,西药组予以赖诺普利处理,中药组予以参草通脉颗粒处理。治疗后PCR法检测心肌组织中MMP-2,TIMP-2,AngⅡ,ACE2的相对表达量。结果中药高剂量组对MMP-2的抑制效果和对TIMP-2的促进效果明显优于西药组,差异有统计学意义(P<0.05);中药组及西药组AngⅡ水平明显低于模型组,差异具有统计学意义(P<0.05),西药组对AngⅡ抑制效果优于中药中剂量组,但与中药高剂量组比较,差异无统计学意义(P>0.05);中药组及西药组ACE2水平显著高于模型组,差异具有统计学意义(P<0.05);其中中药高剂量组和中药中剂量组对ACE2的促进效果均优于西药组,差异有统计学意义(P<0.05),中药高剂量组效果优于中药中剂量组,差异具有统计学意义(P<0.05)。结论参草通脉颗粒可以抑制MMP-2的表达,增加TIMP-2的表达,促进ACE2的表达从而对抗AngⅡ的作用,起到保护心肌,延缓CHF的进展,高剂量的参草通脉颗粒疗效更为明显。Objective To investigate the effects of Shencao Tongmai Granules(参草通脉颗粒)on MMP-2,TIMP-2,AngⅡ,ACE2 and ventricular remodeling in rats with chronic heart failure.Methods Eighty rats were randomly selected as sham operation group(n=15),and 65 models of chronic heart failure were constructed.After successful modeling,44 rats were selected and divided into model group,western medicine group and Chinese medicine high,middle and low dose groups,9 in each group.The model group and the sham operation group were treated with distilled water.The western medicine group was treated with lisinopril,and the Chinese medicine group was treated with Shencao Tongmai Granule.After treatment,the relative expressions of MMP-2,TIMP-2,AngⅡand ACE2 in myocardial tissue was detected by PCR.Results The inhibitory effect of high dose group on MMP-2 and TIMP-2 was significantly better than that of the western medicine group(P<0.05).The level of AngⅡin Chinese medicine group and western medicine group was significantly lower than that in model group.The difference was statistically significant(P<0.05).The western medicine group had better inhibition effect on AngⅡthan the traditional Chinese medicine group,but the difference was not statistically significant(P>0.05).The ACE2 level in the Chinese medicine group and western medicine group was significant.Compared with that of the model group,the difference was statistically significant(P<0.05).Among them,the high and middle dose groups had better promotion effect on ACE2 than the western medicine group.The difference was statistically significant(P<0.05).The effect of the high dose group was better than that of the middle dose group and the difference was statistically significant(P<0.05).Conclusion Shencao Tongmai Granule can inhibit the expression of MMP-2,increase the expression of TIMP-2,promote the expression of ACE2 and counteract the effect of AngⅡ,protect the myocardium and delay the progression of CHF.High dose of Shencao Tongmai Granules has more obvious effect.
