摘  要:目的基于网络药理学探讨水蛭防治股骨头坏死(ONFH)的潜在作用机制。方法应用中药综合数据库(TCMID)及BATMAN-TCM分析工具筛选出单味中药水蛭的活性成分及作用靶点,检索与疾病靶点有关的数据库GeneCadrs、TTD、OMIM筛选ONFH的作用靶点,进而筛选水蛭与ONFH的交集核心靶点,并运用Cytoscape 3.7.1软件构建化合物-靶点-疾病网络,基于DAVID数据库对靶点进行GO功能富集和KEGG通路富集分析。结果基于BATMAN-TCM分析工具预测得出水蛭防治ONFH的活性成分13个和交集靶点28个;GO功能富集分析得到条目153个,其中生物过程条目117个、细胞组成条目10个、分子功能条目26个;KEGG通路富集分析得出条目35个,主要包括PPAR信号通路、TNF信号通路、AMPK信号通路、NF-κB信号通路、TLR信号通路等。结论基于网络药理学初步探讨并验证了水蛭防治ONFH多成分-多靶点-多通路的整体调节作用特点,预测了水蛭防治ONFH的潜在作用机制,以期为其活性成分研究与实验研究提供科学依据。Objective To explore the potential mechanism of leech in the prevention and treatment of osteonecrosis of the femoral head(ONFH)based on network pharmacology.Methods Comprehensive application of traditional Chinese medicine integrative database(TCMID)and BATMAN-TCM analysis tool were used to select the active ingredients and targets of leech,the disease target databases such as GeneCadrs,TDD and OMIM were used for screening ONFH targets,and then we screened leech and the intersection of ONFH core targets,and used Cytoscape 3.7.1 software to build compound-targets-disease network.Based on DAVID database,we carried out GO enrichment analysis and KEGG pathway enrichment analysis.Results Based on BATMAN-TCM analysis,13 active components and 28 intersection targets of leech were predicted.GO functional enrichment analysis obtained 153 items,including 117 biological processes,10 cell components and 26 molecular functions.KEGG pathway enrichment analysis revealed 35 items,including PPAR signaling pathway,TNF signaling pathway,AMPK signaling pathway,NF-κB signaling pathway and TLR signaling pathway.Conclusion Based on the network pharmacology,the overall regulation of using leech to control ONFH multi-component-multi-target-multi-pathway was preliminarily discussed and verified,and the potential mechanism of leech controlling ONFH was predicted,so as to provide scientific basis for the study of its active components and experimental studies.
