60Co-γ射线辐射灭菌对天麻药材指纹图谱及有效成分的影响

摘　　要：目的:建立天麻药材指纹图谱,考察不同剂量60Co-γ射线辐照灭菌对天麻药材中天麻素、对羟基苯甲醇、巴利森苷A、巴利森苷B、柠檬酸5个有效成分含量的影响。方法:利用HPLC法,采用wondasil C18(250 mm×4.6 mm,5μm)液相色谱柱,以乙腈-0.1%磷酸溶液为流动相,梯度洗脱;流速为1.0 ml·min^(-1);检测波长为220 nm;柱温为30℃,进样量为10μl,建立天麻药材HPLC指纹图谱。分别选择辐照剂量2,6,10 k Gy对天麻进行辐照,比较辐照前后天麻指纹图谱及5个有效成分的变化,并采用SPSS 22.0软件分析5个有效成分辐照前后含量变化。结果:共确定14个共有峰,不同剂量辐照前后天麻药材指纹图谱的相似度为0.918~0.999;辐照剂量不超过6 k Gy时,天麻药材中的天麻素、对羟基苯甲醇、巴利森苷A、巴利森苷B、柠檬酸含量变化无统计学意义(P>0.05)。结论:所建立的天麻指纹图谱及5个成分含量测定方法可作为天麻质控方法,在辐照剂量不超过6 k Gy时,60Co-γ辐照灭菌法可用于天麻药材的灭菌。Objective: To establish the fingerprint of Gastrodia Rhizoma,investigate the influence of different doses of 60Co-γ ray irradiation on the five effective components in Gastrodia Rhizoma including gastrodin,4-hydroxybenzyl alcohol,parishin A,parishin B and citric acid.Methods: The HPLC analysis was performed on a Wondasil C18(250 mm×4.6 mm,5 μm) column with acetonitrile-0.1% phosphoric acid as the mobile phase in a gradient elution mode,the flow rate was 1.0 ml·min^(-1),the detection wavelength was 220 nm,the column temperature was 30 ℃,and the sample size was 10 μl.The fingerprints and five active components of Gastrodia Rhizoma were compared before and after irradiation.The contents of five active components were analyzed by SPSS 22.0 software.Results: The fingerprint was established with 14 common peaks,the similarity was within the range of 0.918-0.999;when the radiation dose was lower than 6 k Gy,the contents of gastrodin,4-hydroxybenzyl alcohol,parishin A,parishin B and citric acid had no statistically significant differences(P>0.05).Conclusion: The established method can be used for the quality control of Gastrodia Rhizoma.When the radiation dose is not more than 6 kGy,60Co-γ ray irradiation can be used for the sterilization of Gastrodia Rhizoma.
