摘 要:目的建立淫羊藿药材中5种成分的高效液相色谱法,采用双标线性校正法计算保留时间,用于色谱峰的定性分析。方法选用13根不同品牌的C_(18)色谱柱,测定淫羊藿药材中5种成分的实际保留时间,计算平均值得到标准保留时间(tSRT),以峰2朝藿定B和峰5宝藿苷I 2个成分作为双标化合物,使用双标线性校正法预测保留时间,并在4根其他品牌的C_(18)色谱柱上进行了验证。同时以淫羊藿苷为参照物,计算校正因子,用相对保留时间法预测保留时间。比较双标线性校正法与相对保留时间法的优劣。结果与相对保留时间法相比较,双标线性校正法具有更高的色谱柱符合率和保留时间预测正确率,明显优于相对保留时间法。结论双标线性校正法能更好的辅助色谱峰定性,是一种新的对照品替代方法,具有较好的应用前景。OBJECTIVE To establish a method for identification of five components of Herba Epimedii by HPLC chromatographic for the qualitative peak analysis.METHODS The retention time of the five components in Herba Epimedii was determined on 13 C_(18) columns of different brands.The standard retention time(SRT)was calculated.Both epime B(peak 2)and baohuoside I(peak 5)were taken as the linear calibration standards to forecast the retention time of the other three components.Other four C_(18) columns of different brands were used for validation.In comparison of liner calibration with the two reference substances,icariin was used as the reference substance for the determination of relative correction factors of the other four components.RESULTS Liner calibration with two reference substances was more accurate for predicting the retention time and suitable for more kinds of chromatographic columns than the relative retention time method.CONCLUSION The method of liner calibration with two reference substances can be used to assist in the chromatographic peak identification,with a broad practical prospect.
