摘  要:目的观察眼针对急性脑缺血再灌注损伤(CIRI)模型大鼠脑源性神经因子(brain-derived neurotrophic factor,BDNF)表达的影响及环磷酸腺苷反应元件结合蛋白(cAMP response element binding protein,CREB),探讨眼针治疗CIRI的机制。方法将40只SPF级雄性SD大鼠按照数字表法随机分为5组:空白对照组、假手术组、模型组、眼针组、非穴区组,每组8只。采用线栓法制备大鼠右侧大脑中动脉缺血再灌注损伤模型。于再灌注后24 h采用ZeaLonga评分法对大鼠进行神经功能缺损评分。采用ELISA方法测定脑组织中BDNF蛋白的含量,采用Western Blot方法检测脑组织中p-CREB、CREB蛋白水平。结果与模型组比较,眼针组大鼠神经功能缺损评分明显下降;BDNF蛋白含量明显升高(P<0.05);脑组织p-CREB和CREB蛋白水平明显上调(P<0.05)。非穴区组与神经功能缺损评分与模型组相近,BDNF、p-CREB和CREB蛋白水平与模型组无明显差异(P>0.05),无统计学意义。结论眼针能改善大鼠急性脑缺血再灌注损伤;其机制可能是通过眼针促进BDNF产生,增加神经元修复;促进p-CREB蛋白表达,减少神经细胞凋亡,进而发挥神经保护作用从而减轻脑缺血损伤。Objective To study the effects of eye acupuncture therapy on expressions of BDNF and CREB in rats with acute cerebral ischemia reperfusion injury and the related mechanism.Methods Forty SPF male SD rats were randomly divided into five groups according to digital table method:blank control group,sham operation group,model group,eye acupuncture group and non-point area group,with 8 rats in each group.Rat models of ischemia reperfusion of right middle cerebral artery occlusion were established by the suture method.After reperfusion for 24 hours,ZeaLonga score was used to evaluate the neurological deficit in rats.The content of BDNF protein in rat brains was determined by ELISA,and the levels of p-CREB and CREB protein in brains were detected by Western Blot.Results Compared with the model group,the scores of neurological impairment in the eye acupuncture group were significantly decreased,the expression of BDNF protein was significantly increased(P<0.05),and the levels of p-CREB and CREB protein in brain tissue were significantly increased(P<0.05).There was no significant difference in the levels of BDNF,p-CREB or CREB protein between non-point area group and model group(P>0.05).Conclusion Eye acupuncture can improve the acute cerebral ischemia reperfusion injury in rats.The mechanism may be that eye acupuncture therapy can promote the production of BDNF,increase the repair of neurons,promote the expression of p-CREB protein,reduce the apoptosis of neurons,and then play a neuroprotective role to alleviate cerebral ischemia reperfusion injury.
