摘  要:目的研究参草通脉颗粒对慢性心力衰竭大鼠心肌细胞ACE 2、MMP 2、TIMP 2的影响。方法取1~3 d的Wistar大鼠心脏并剪成1 mm3大小的组织块,经消化、振荡制成细胞悬液,离心分离心肌细胞,计数后接种于培养皿。将培养皿中心肌细胞加入6孔板,应用AngⅡ诱导心肌细胞。将36只Wister大鼠随机分为正常组、模型组、西药组和中药高、中、低剂量组,每组6只。正常组大鼠常规等容积蒸馏水灌胃,西药及中药组大鼠按人鼠剂量换算给予药物灌胃,用药5 d后心脏采血,自然分离血清。按组别加入AngⅡ诱导的心衰心肌细胞中。培养36 h后,Elisa法检测各组心肌细胞氨基末端脑钠尿肽(NT-proBNP)水平,Western Blot法检测各组心肌细胞基质金属蛋白酶2(MMP 2)、基质金属蛋白酶组织抑制因子2(TIMP 2)、血管紧张素转化酶2(ACE 2)蛋白含量,实时荧光定量法(PCR)检测MMP 2 mRNA、TIMP 2 mRNA、ACE 2 mRNA表达。结果与正常组比较,模型组NT-proBNP、MMP 2蛋白含量、MMP 2 mRNA表达显著升高,TIMP 2、ACE 2蛋白含量及TIMP 2、ACE 2 mRNA表达显著降低(P<0.05);与模型组比较,西药组、中药组NT-proBNP、MMP 2蛋白含量、MMP 2 mRNA表达显著降低,TIMP 2、ACE 2蛋白含量及TIMP 2、ACE 2 mRNA表达显著升高(P<0.05),具有统计学意义;与西药组比较,中药组上述指标差异无统计学意义(P>0.05)。结论参草通脉颗粒能调节AngⅡ诱导的大鼠心衰心肌细胞中MMP 2、TIMP 2、ACE 2表达。Objective To study the effects of Shencao Tongmai Granule(参草通脉颗粒)on ACE 2,MMP 2 and TIMP 2 in myocardial cells of chronic heart failure rats.Method We took the heart out of 1~3 days old Wistar rats and cut it into 1 mm3 size tissue block.After digesting and shaking,we made the cell suspension,centrifuged to separate cardiomyocytes,and then counted and inoculated the tissue in culture dish.Cardiomyocytes in the culture dish were added to a 6-well plate,and cardiomyocytes were induced using AngⅡ.Thirty-six Wister rats were randomly divided into normal group,model group,Western medicine group and traditional Chinese medicine high,medium and low dose groups,with 6 rats in each group.The rats in the normal group were intragastrically administrated with equal volume of distilled water.The rats in the Western medicine and the traditional Chinese medicine group were administered with the drug according to the dose of human and mice.After 5 days of treatment,the blood was collected from the heart and the serum was naturally separated.AngⅡ-induced heart failure cardiomyocytes were added by groups.After 36 hours of culture,the levels of NT-proBNP in cardiomyocytes of each group were detected by ELISA method.The contents of MMP 2,TIMP 2 and ACE 2 protein were detected by Western Blot.The expressions of MMP 2 mRNA,TIMP 2 mRNA and ACE 2 mRNA were detected by PCR.Results Compared with the normal group,NT-proBNP,MMP 2 protein content and MMP 2 mRNA expression were significantly increased in the model group.Protein contents of TIMP 2 and ACE 2 and expressions of TIMP 2 and ACE 2 mRNA were significantly decreased(P<0.05).Compared with Western medicine group and traditional Chinese medicine group,protein contents of NT-proBNP and MMP 2 and MMP 2 mRNA expression were significantly decreased.Protein contents of TIMP 2 and ACE 2 and expressions of TIMP 2 and ACE 2 mRNA were significantly increased(P<0.05).Compared with the Western medicine group,there was no significant difference of the above indicators in the traditi
