摘 要:目的基于网络药理学探讨肉苁蓉治疗血管性痴呆(vascular dementia,VD)的作用机制。方法采用TCMSP分析平台检索中药活性成分和中药靶点,利用GeneCards数据库和OMIM数据库筛选肉苁蓉治疗VD的作用靶点,构建蛋白互作网络(PPI),进行GO功能富集分析及KEGG通路富集分析,构建可视化的中药-疾病靶点相关的信号通路。结果从肉苁蓉中筛选出4个有效成分,作用于60个血管性痴呆靶点,肉苁蓉治疗VD的核心基因主要有IL6、VEGFA、EGFR、CASP3、MYC、CCND1、FOS、PPARG、AR、RELA、NOS3、ICAM1等。中药-疾病靶点涉及的GO功能主要包括DNA结合转录激活活性、核受体活性、转录因子活性,直接配体调节的序列特异性DNA、乙酰胆碱受体活性、泛素蛋白连接酶结合、RNA聚合酶II转录因子结合等。中药-疾病靶标涉及的KEGG通路主要包括流体剪切应力与动脉粥样硬化信号通路、前列腺癌信号通路、糖尿病并发症中AGE-RAGE信号通路、肿瘤坏死因子信号通路、癌症中的蛋白多糖信号通路、PI3K-Akt信号通路、肝细胞癌信号通路、细胞凋亡信号通路、丙肝信号通路等。结论基于网络药理学探讨了肉苁蓉治疗血管性痴呆的分子机制,为中药现代化研究提供了理论基础。Objective Based on the network pharmacology to study the mechanism of Roucongrong(Cistanches Herba)in the treatment of vascular dementia(VD).Methods By using TCMSP analysis platform,we retrieved active ingredients and targets in traditional Chinese medicine(TCM).GeneCards database and OMIM database were used for screening targets of Roucongrong(Cistanches Herba)treating VD to construct the protein-protein interaction network(PPI).GO function enrichment analysis and KEGG pathway enrichment analysis were used to build the visualization of traditional Chinese medicine-disease targets related signaling pathway.Results There were four effective constituents screened out from Roucongrong(Cistanches Herba)and 60 vascular dementia targets.The core genes for the treatment of VD were mainly IL6,VEGFA,EGFR,CASP3,MYC,CCND1,FOS,PPARG,AR,RELA,NOS3,ICAM1,etc.GO function of traditional Chinese medicine(TCM)-disease targets mainly included DNA-binding transcription activation activity and nuclear receptor activity and the activity of transcription factors,ligand directly regulate DNA sequence specificity,acetylcholine receptor activity and protein ubiquitin ligase combination and RNA polymerase II transcription factor binding.KEGG pathways of Traditional Chinese medicine(TCM)-disease targets mainly included fluid shear stress and atherosclerosis signaling pathways,signaling pathways of prostate cancer,diabetes complications in AGE-RAGE signaling pathways,tumor necrosis factor signaling pathways,polysaccharide protein signaling pathways in cancer,PI3 K-Akt signaling pathways,hepatocellular carcinoma(HCC)signal pathway,cell apoptosis signaling pathways and hepatitis C signaling pathways.Conclusion Based on the network pharmacology,the molecular mechanism of Roucongrong(Cistanches Herba)in the treatment of vascular dementia was explored,providing a theoretical basis for the research of TCM modernization.
