摘  要:为了探究SPIA1基因的表达量与蛋壳绿色深浅之间的规律性,本试验以赤水乌骨鸡母鸡为试验动物,利用双重PCR技术进行基因分型,根据基因型(纯合型、杂合型)和蛋壳颜色(深绿色、浅绿色、淡绿色)进行分类。采集300日龄赤水乌骨鸡的蛋壳腺进行转录组测序,结果发现SPIA1基因为蛋壳绿色的上调基因,为了验证测序结果,随机选取不同基因型不同蛋壳颜色不同产蛋期的赤水乌骨鸡母鸡各3只,共54只进行屠宰,收集肝脏和蛋壳腺,提取总RNA,逆转录成cDNA后进行qPCR反应,检测SPIA1基因在组织中的表达量。结果显示:随着蛋壳绿色加深,蛋壳腺和肝脏中SPIA1基因的表达量增高,纯合子中的表达量高于杂合子,并且不同的产蛋期,都表现为蛋壳腺的表达量高于肝脏的表达量。研究结果为绿壳性状的遗传机制研究提供基础资料。To identify the relationship between SPIA1 gene expression and eggshell color depth,Chishui silky fowls(hens)at 180 days of age,360 days of age,and 540 days were used as experimental subjects.Duplex PCR was used to perform genotyping for the hens.Transcriptome sequencing was performed on the eggshell glands of the hens at 300 days of age.Analysis and sequencing results showed that SPIA1 was the up-regulating gene of the color green of the eggshell.To verify the sequencing results,three hens were chosen randomly from each type of the chickens based their different genotypes,different depths of the color green of eggshell,and different egglaying periods.And shell gland and liver samples were collected these hens.Next,total RNA was extracted from the birds and was reversely transcribed into cDNA.Finally,the expressions of the SPIA1 gene in different tissues were detected using Quantitative Real-time PCR.The results showed that the expression of SPIA1 in the eggshell gland and liver of the Chishui silky fowl increased along with the increase of eggshell green.The expression level in their homozygote was higher than that in their heterozygote,which was consistent with the results of related analyses.This study provided basic information for research on the genetic mechanism of the green shell traits of silky fowl.
