摘 要:目的探讨黄芪多糖对苯诱导小鼠白细胞减少症的影响。方法苯和玉米油按照1∶1配制混合液,按照苯含量1.3g·kg^(-1)灌胃制作白细胞减少的小鼠模型。将小鼠随机分为6组:空白组、模型组、黄芪多糖低、中、高剂量实验组(100,200,400 mg·kg^(-1)黄芪多糖)和阳性对照组(100 mg·kg^(-1)地榆升白片),每组12只。空白组给与等体积的玉米油灌胃,其余各组均采用苯和玉米油按照苯含量1.3 g·kg^(-1)灌胃,连续给药4周。用血球计数板计数骨髓有核细胞数;用流式细胞仪检测CD34+细胞的百分率;用Bauin染色计数脾集落生成单位(CFU-S);用称重法计算小鼠的脾和胸腺的脏器指数,用酶联免疫试剂盒测定血清粒细胞集落刺激因子(G-CSF)的数量。结果给药后14 d,空白组、模型组和高剂量实验组的白细胞(white blood cell,WBC)分别为(5.32±1.01)×10^(9)·L^(-1),(1.73±0.48)×10^(9)·L^(-1)和(5.87±1.11)×10^(9)·L^(-1);给药后28 d,空白组、模型组和高剂量实验组的WBC分别为(5.24±0.76)×10^(9)·L^(-1),(1.12±0.43)×10^(9)·L^(-1)和(6.6±0.91)×10^(9)·L^(-1);给药后,空白组、模型组和高剂量实验组的骨髓有核细胞数分别为(35.82±6.91)×106·L^(-1),(9.62±1.34)×106·L^(-1)和(35.93±3.78)×106·L^(-1);空白组、模型组和高剂量实验组CFU-S水平分别为(58.74±1.21)pg·mL^(-1),(25.78±2.30)pg·mL^(-1),(56.85±2.83)pg·mL^(-1);空白组、模型组和高剂量实验组的CD34+细胞百分比分别为(46.34±0.92)%,(27.43±1.48)%和(58.92±1.74)%;空白组、模型组和高剂量实验组的血清G-CSF水平分别为(62.44±1.02),(26.88±1.82)和(59.74±1.89)pg·mL^(-1);模型组与空白组比较或者高剂量实验组与模型组比较,上述指标的差异具有统计学意义(均P<0.05)。结论黄芪多糖可促进小鼠白细胞含量、骨髓有核细胞、CFU-S、血清G-CSF水平及脾和胸腺的脏器指数等的升高,对白细胞减少有治疗作用。Objective To investigate the effect of astragalus polysaccharide(APS)on benzene-induced leukopenia in mice.Methods The mixture of benzene and corn oil was prepared in 1∶1,and the mouse model of leukopenia was made by gavage with benzene content of 1.3 g·kg^(-1).Mice were randomly divided into 6 groups:blank group,model group,low,medium and high-dose APS experimental group(100,200,400 mg·kg^(-1)APS)and positive control group(100 mg·kg^(-1)Diyushengbai tablet),with 12 mice in each group.The blank group was given corn oil of the same volume by gavage,and the other groups were given by mixture of benzene and corn oil with benzene content of 1.3 g·kg^(-1)by gavage for 4 weeks continuously.The number of nucleated cells in bone marrow was counted by blood cell counting board.The percentage of CD34+cells was determined by flow cytometry.Colony forming unit of spleen(CFU-s)is counted with Bauin staining;The viscera indices of the spleen and thymus of mice were measured by weighing,and the quantity of granulocyte colony-stimulating factor(G-CSF)was measured with El ISA kit.Results On 14 days after administration,white blood cells(WBC)in the blank group,model group and high-dose experimental group were(5.32±1.01)×10^(9)·L^(-1),(1.73±0.48)×10^(9)·L^(-1)and(5.87±1.11)×10^(9)·L^(-1),respectively.On day 28 after administration,WBC of the blank group,model group and high-dose experimental group was(5.24±0.76)×10^(9)·L^(-1),(1.12±0.43)×10^(9)·L^(-1)and(6.16±0.91)×10^(9)·L^(-1),respectively.After administration,the number of myeloid nucleated cells in the blank group,the model group and the high-dose experimental group were(35.82±6.91)×106·L^(-1),(9.62±1.34)×106·L^(-1)and(35.93±3.78)×106·L^(-1),respectively.The CFU-S levels in the blank group,the model group and the high-dose experimental group were(58.74±1.21)pg·mL^(-1),(25.78±2.30)pg·mL^(-1)and(56.85±2.83)pg·mL^(-1),respectively.The percentages of CD34+cells in the blank group,model group and high-dose experimental group were(46.34±0.92)%,
