摘 要:目的研究沉默乳腺癌扩增序列2(BCAS2)基因对乳腺癌MDA-MB-231细胞凋亡的影响。方法将细胞分3组:空白组、对照组和实验组。空白组细胞不做任何处理,对照组细胞转染空载体(3×10^(8)TU·mL^(-1),10μL),实验组细胞转染慢病毒载体pGLV3-GFP-shBCAS2(3×10^(8)TU·mL^(-1),10μL)。用AnnexinV-FITC/PI双染法检测细胞凋亡率;用试剂盒法检检测Caspase-3活性(光密度值);蛋白质印迹法检测caspase-3蛋白表达(灰度值)。结果空白组、对照组和实验组的细胞凋亡率分别为(12.33±1.08)%,(12.56±0.77)%和(44.93±3.22)%;这3组的Caspase-3活性分别为1.00±0.06,1.02±0.07和3.91±0.70;这3组的caspase-3蛋白相对表达量分别为0.21±0.02,0.19±0.03和0.42±0.04。实验组与空白组和对照组相比,上述指标的差异均有统计学意义(均P<0.01)。结论沉默BCAS2基因可诱导乳腺癌MDA-MB-231细胞凋亡,其机制可能与caspase-3激活有关。Objective To investigate the effect of breast cancer amplified sequence 2(BCAS2)silence on apoptosis in breast cancer MDA-MB-231 cells.Methods Cells were divided into three groups:blank group,control group and experimental group.The blank group was transfected nothing,the control group was transfected with control vector(3×10^(8)TU·mL^(-1),10μL),while the experimental group was transfected with lentivirus vector shBCAS2(3×10^(8)TU·mL^(-1),10μL).The cell apoptosis was detected by Annexinv-FITC/PI double staining.The activity(optical density)of caspase-3 was assessed by detection kit.The protein level(gray value)of caspase-3 was detected by Western blot.Results The apoptosis rates in the blank,control and experimental groups were(12.33±1.08)%,(12.56±0.77)%and(44.93±3.22)%,respectively;the activity of capase-3 in the three groups were 1.00±0.06,1.02±0.07 and 3.91±0.70,respectively;the protein expressions of caspase-3 in the three groups were 0.21±0.02,0.19±0.03 and 0.42±0.04.Comparison between experimental group and blank or control group,the difference of the factors were significant(all P<0.01).Conclusion Silencing of BCAS2 can promote apoptosis of breast cancer MDA-MB-231 cells,which might be related to activation of caspase-3.
