泡球蚴感染小鼠肝脏差异表达miRNA筛选及其作用通路分析

摘  要:目的使用微小RNA(miRNA)芯片筛选泡球蚴感染小鼠3个月的肝脏和对照未感染小鼠肝脏的差异miRNA,探讨其下游靶基因及相关调控通路在肝泡型包虫病中可能的生物学功能。方法雌性Balb/c小鼠肝脏穿刺注射泡球蚴,感染3个月后,以肝脏成功寄生泡球蚴的小鼠为感染组,对照组为同期腹腔注射等容量生理盐水,正常组为健康雌性Balb/c小鼠。采集各组小鼠的肝脏组织,采用HE染色法检测各实验组的肝脏病理。采用miRNA微阵列芯片检测感染组和对照组肝脏中的miRNA表达谱。使用TargetScan数据库预测差异表达miRNAs的靶基因,利用David数据库分析其靶基因的生物学功能和信号通路。结果病理染色结果显示感染组小鼠肝脏成功寄生泡球蚴。基因芯片结果提示,与未感染组小鼠相比,感染组小鼠肝脏中有46个miRNAs具有差异,其中29个上调和17个下调。生物信息学分析显示,差异表达miRNA靶基因与细胞过程、生物调节、转录活性调节等重要细胞生物功能相关,涉及轴突引导、转化生长因子-β(TGF-β)和胞吞作用等通路。结论泡球蚴感染导致小鼠肝脏miRNAs表达谱发生变化,提示miRNAs可能参与了泡球蚴的致病过程。Objective The aim of this study was to screen the differentially expressed miRNAs in the liver of mice infected with the larvae of Echinococcusmultilocularis and non-infected mice,and explore the biological role of their downstream target genes and related regulatory pathways.Methods Female Balb/c mice were randomly divided into sham-operated group(the saline group),the model group(E.multilocularisinfected group)and healthy control group.Three months after infection,the liver pathology of each group was detected by HE staining.The miRNA expression profile was detected by miRNA microarray.The target genes of differentially expressed miRNAs and their biological functions and signaling pathways were predicted by Targetscan and David databases.Results A total of 46 differentially expressed miRNAs were identified between the model group and the saline group,among which 29 were up-regulated and 17 were down-regulated.Bioinformatics analysis shows that differentially expressed miRNA target genes were related to cell processes,biological regulation,transcriptional activity regulation and other important cellular biological functions,and related to neural guidance,TGFβsignaling pathway and endocytosis signaling pathway.Conclusion The expression of miRNAs in the liver of mice infected with E.multilocularis is significantly changed,suggesting that miRNAs play an important role in the pathogenesis of alveolar echinococcosis.
