摘 要:目的研究金银花药材、标准汤剂、中间体、配方颗粒特征图谱的相关性。方法采用HPLC法进行研究,使用Thermo Scientific AcclaimTM120 C18色谱柱(5μm,4.6×250 mm);以乙腈-0.4%磷酸水溶液为流动相进行梯度洗脱;检测波长为327 nm;柱温为30℃;流速为1 mL/min;进样量为10μL。比较15批金银花药材、标准汤剂、中间体、配方颗粒特征图谱及相关性。结果金银花药材、标准汤剂、中间体、配方颗粒特征图谱均可指认出新绿原酸、绿原酸、隐绿原酸、异绿原酸B、异绿原酸A、异绿原酸C等6个成分。15批金银花药材、标准汤剂、中间体、配方颗粒各特征图谱相似度均大于0.9。金银花药材、标准汤剂、中间体、配方颗粒中主要化学成分组成基本相同。结论建立的HPLC法特征图谱方法可用于金银花药材、标准汤剂、中间体、配方颗粒的质量控制。Objective To study the correlation of characteristic chromatograms of pieces,standard decoction,intermediate and formula granule of Lonicera japonica flos.Methods The results were studied by HPLC method;column was Thermo Scientific AcclaimTM120 C18(5μm,4.6×250 mm);acetonitrile-0.4%phosphoric acid aqueous solution was used as mobile phase for gradient elution.The detection wavelength was 327 nm.The column temperature was 30℃.The flow rate was 1 mL/min.The injection volume was 10μL.Characteristic chromatograms and correlation with 15 batches of pieces,standard decoction,intermediate and formula granule of Lonicera japonica flos were compared.Results Six components,including neochlorogenic acid,chlorogenic acid,crypto-chlorogenic acid,isochlorogenic acid B,isochlorogenic acid A and isochlorogenic acid C,were identified by the characteristic chromatograms for 15 batches of pieces,standard decoction,intermediate and formula granule of Lonicera japonica flos.The similarity of characteristic chromatograms of 15 batches of pieces,standard decoction,intermediate and formula granule of Lonicera japonica flos were all greater than 0.9.The main chemical components of pieces,standard decoction,intermediate and formula granule of Lonicera japonica flos were basically the same.Conclusion The established HPLC characteristic chromatograms method can be used for the quality control of pieces,standard decoction,intermediate and formula granule of Lonicera japonica flos.
