摘 要:目的基于不同物体认知实验模式开展慢性睡眠干扰引起的学习记忆功能研究,为航天医学相关药物筛选提供行为学检测方法参考。方法48只雄性小鼠随机分为3批,每批对照组和模型组各8只;模型组小鼠采用滚筒法造模模拟睡眠干扰,滚筒设置参数为:60 s/圈,转1圈,间隔2 min;连续造模14天后分别对3批动物进行时序记忆实验、情景记忆实验以及新物体识别实验,以检测小鼠学习记忆能力。结果在时序记忆和情景记忆实验中,与对照组相比,模型组小鼠的相对辨别指数显著降低(P<0.05,P<0.01)。在新物体识别实验中,模型组小鼠相对辨别指数在不同时间段均降低(P<0.05,P<0.01)。结论慢性睡眠干扰可损害小鼠的物体认知时序记忆、情景记忆和新物体识别能力。Objective The learning and memory dysfunction induced by chronic sleep interruption was studied with different object recognition tests to provide a behavioral test method for screening space medicine related drugs.Methods Forty-eight male mice were randomly divided into three batches and each batch were randomly divided into the control group and the model group with eight animals in each group.Mice in the model group were treated with rotating sleep interferometer for 14 days(The rotation speed was 60 s one rotation and there was a 2 min pause after each rotation).Then the temporal order memory task,the object context recognition task,and the novel object recognition task were tested for the three batches of mice respectively to assess the recognition behavioral activities of mice.Results For the temporal order memory task and object context recognition task,there was a significant decrease in discrimination index in the model group as compared with the control group(P<0.05,P<0.01).For the novel object recognition task,the discrimination index significantly decreased in the model group at different time phases(P <0.05,P<0.01).Conclusion Chronic sleep interruption could impair the object recognition memory,the temporal memory and object context memory in mice.
