摘  要:目的建立15批满山红叶HPLC指纹图谱,测定已知成分杜鹃素含量。方法运用高效液相色谱法建立满山红叶指纹图谱并测定杜鹃素的含量,色谱条件:Shim-Pack XR-ODSⅢ(2.0 mmi.d×150 mm)色谱柱,流速:0.2 mL·min^(-1),柱温:25℃,流动相甲醇-0.3%甲酸水溶液梯度洗脱,检测波长297 nm;分别进行15批满山红叶指纹图谱相似度评价和聚类分析。结果建立了满山红叶指纹图谱,有10个共有峰,从中找到了已知成分杜鹃素,其含量在0.011%~0.016%,15批满山红叶指纹图谱整体相似度>0.9,具有良好的相似度。聚类分析结果显示,将15批满山红叶共分为4类。结论满山红叶指纹图谱较好的反映了满山红叶多组分的特征,所选满山红药材在产地和批次上具有代表性,为后续满山红药材的研究提供参考。Objective To establish HPLC fingerprints of 15 batches of rhododendron leaves and determine the content of known component of farrerol.Methods The fingerprints of rhododendron were established by high performance liquid chromatography(HPLC)method and the content of farrerol was determined.The chromatographic conditions were as follows:Shim-Pack XR-ODSⅢ(2.0 mmi.d×150 mm)column,maintained at 25℃and eluted with acetonitrile-0.3%formic acid at a flow rate of 0.2 ml/min and with the detection wavelength at 297 nm.Similarity evaluation and cluster analysis of the fingerprints were carried out by Similarity Evaluation for Fingerprint of TCM(Version 2012)and the software IBM SPSS Statistic(Version 20.0).Results The fingerprints of rhododendron leaves were established with 10 common peaks,and the known component of farrerol was found,with a content between 0.011%and 0.016%.The overall similarity of 15 batches of rhododendron leave fingerprints was more than 0.9,showing a good similarity.Cluster analysis showed that 15 batches of rhododendron leaves could be divided into 4 groups.Conclusion The fingerprints of rhododendron better reflect its multi-component characteristics.The selected rhododendron medicinal materials are representative in origin and batch,which can provide a reference for subsequent research on this kind of medicinal materials.
