摘  要:目的:观察氟化钠对大鼠心肌细胞中自噬相关基因微管相关蛋白1轻链3(LC-3)、哺乳动物ATG6同源蛋白(Beclin-1)、信号转导与转录激活因子3(STAT-3)基因表达的影响。方法:将大鼠H9c2心肌细胞加入不同浓度氟化钠(NaF)的培养基,培养48 h后提取心肌细胞的RNA,RT-PCR法比较各组心肌细胞中LC-3、STAT-3、Beclin-1 mRNA的表达情况。结果:48 h后,与对照组相比,加入2.5 mg/L、5 mg/L、10 mg/L、20 mg/L、40 mg/L NaF的心肌细胞中LC-3、Beclin-1的表达量明显增高(P<0.05),STAT-3相对表达量降低(P<0.05)。结论:心肌细胞LC-3和Beclin-1 mRNA表达量随NaF浓度的增加而增加,STAT-3 mRNA表达量随NaF浓度的增加而减少,提示氟中毒导致心肌细胞活力降低可能与自噬的过度激活有关。Objective:To observe the effects of sodium fluoride on autophagy-related gene microtubule-associated protein 1 light chain 3(LC-3),mammalian Atg6 homologous protein autophagy(Beclin-1),signal transducer and activator of transcription 3(STAT-3)genes in rat cardiomyocytes.Methods:Rat H9c2 cardiomyocytes were added with media containing different concentrations of sodium fluoride(NaF),and the cardiomyocyte RNA was extracted after 48 hours’culturing.The expression of LC-3,STAT-3,and Beclin-1 mRNA in each group of cardiomyocytes were compared by RT-PCR.Results:After 48 hours,compared with the control group,the expression levels of LC-3 and Beclin-1 in cardiomyocytes added with 2.5 mg/L,5 mg/L,10 mg/L,20 mg/L,and 40 mg/L NaF increased significantly(P<0.05),and the expression level of STAT-3 decreased(P<0.05).Conclusion:The expression of LC-3 and Beclin1 mRNA in cardiomyocytes increases with the increase of NaF concentration,and the expression of STAT-3 mRNA decreases with the increase of NaF concentration,revealing that the damage of cardiomyocytes caused by fluorosis may be related to hyperactivation of autophagy.
