摘 要:目的:比较人骨肉瘤细胞OS-732、Saos-2和U2OS在基质胶(Martrigel)辅助条件下的裸鼠皮下成瘤情况,为研究临床抗肿瘤药物的动物模型奠定基础。方法:利用CCK8实验比较OS-732、Saos-2和U2OS三种骨肉瘤细胞的增殖情况;利用划痕愈合实验比较OS-732、Saos-2和U2OS三种骨肉瘤细胞的迁移情况;利用裸鼠皮下成瘤实验比较OS-732、Saos-2和U2OS三种骨肉瘤细胞移植裸鼠皮下的成瘤情况。将30只裸鼠按照简单随机方法分为6组:OS-732细胞分为A1组(高细胞量)、A2组(低细胞量),Saos-2细胞分为B1组(高细胞量)、B2组(低细胞量),U2OS细胞分为C1组(高细胞量)、C2组(低细胞量)。将各组细胞与Matrigel混悬后,于裸鼠左侧前肢腋下进行皮下注射,观察比较各组成瘤时间、成瘤率和肿瘤大小,并对肿瘤及主要内脏器官进行组织形态分析。结果:A1组和A2组成瘤时间为7天左右,成瘤率均为100%。A1组肿瘤体积和重量显著大于A2组,肿瘤体积分别为[(2167.96±844.85)vs(975.05±987.33)]mm 3,差异有统计学意义(P<0.05);肿瘤质量分别为[(1.33±0.40)vs(0.55±0.50)]g,差异有统计学意义(P<0.01)。B1组和B2组成瘤时间为14天左右,成瘤率分别为100%和20%。肿瘤体积分别为[(1222.67±1171.80)vs 510.86]mm 3;肿瘤质量分别为[(0.75±0.40)vs 0.4]g。由于B2组只有一只小鼠成瘤,因此没有对B1组和B2两组进行统计学比较。C1组和C2组均未能成瘤。结论:Matrigel作为辅助试剂的情况下,裸鼠的成瘤时间和成瘤率与骨肉瘤细胞恶性程度密切相关,同一种骨肉瘤细胞株移植的细胞数量越多,裸鼠成瘤率越高。Objective:To compare the subcutaneous tumorigenesis of human osteosarcoma cells OS-732,Saos-2 and U2OS in nude mice with cell suspension including Matrigel,so as to provide a basis for the study of clinical antitumor drugs in animal models.Methods:CCK8 assay was used to compare the proliferation of OS-732,Saos-2 and U2OS.Scratch healing assay was used to compare the migration of the three osteosarcoma cells.30 nude mice were divided into six groups according to a simple randomization method:OS-732 cells were randomly divided into A1 group(high cell mass),A2 group(low cell mass),Saos-2 cells into B1 group(high cell mass),B2 group(low cell mass),and U2OS cells into C1 group(high cell mass),C2 group(low cell mass).The cells of each group were mixed with Matrigel and injected subcutaneously in the axilla of the left forelimb of nude mice,and the time,rate and size of tumor formation were observed and compared.Results:The tumor formation time of group A1 and group A2 was about 7 days,and the tumor formation rate was 100%.The tumor volume and weight in group A1 were significantly larger than that in group A2,with tumor volume of[(2167.96±844.85)vs(975.05±987.33)]mm 3,respectively,and the difference was statistically significant(P<0.05).The tumor mass was[(1.33±0.40)vs(0.55±0.50)]g,and the difference was statistically significant(P<0.01).The tumor formation time of group B1 and B2 was about 14 days,and the tumor formation rate was 100%and 20%respectively.The tumor volume was[(1222.67±1171.80)vs 510.86]mm 3,respectively.The tumor mass was[(0.75±0.40)vs 0.4]g,respectively.Since only one mouse in group B2 had tumor formation,no statistical comparison was made between group B1 and group B2.Tumor formation was not observed in group C1 and group C2.Conclusion:Matrigel can promote the tumorigenesis of OS-732 and Saos-2 cells in nude mice.The number of transplanted cells of the same osteosarcoma cell line has certain influence on the tumorigenesis of nude mice,but the tumorigenesis rate of nude mice is more related to the
