摘 要:目的探讨以可溶性鸡蛋壳膜蛋白(soluble eggshell membrane protein,SEP)和聚丁二酸丁二醇酯(poly butylene succinate,PBS)共同静电纺丝负载凝血酶制备新型止血敷料的可行性和有效性。方法以PBS/SEP制备新型止血敷料,通过大鼠肝脏创伤模型止血实验、髂动脉出血、肝脏止血材料植入和犬腹主动脉切开缝合后吻合口渗血模型止血实验评估其止血性能及生物安全性;选用人胚纤维母细胞,通过四唑盐比色法试验对其细胞体外毒性进行评价。结果PBS/SEP在大鼠肝脏创伤模型的止血时间和髂动脉出血实验的效果优于对照组,但犬腹主动脉渗血模型的止血时间略长,差异有统计学意义(P<0.05)。细胞毒性检测发现,不同浓度PBS/SEP浸提液在不同时间段的细胞毒性分级均为0~1级。体内实验提示,植入大鼠肝脏后可能部分影响肝功能,对机体肾功能无损害,且不引起机体全身免疫和炎症反应。结论PBS/SEP共同静电纺丝负载凝血酶止血材料是一种有效的止血材料,生物相容性良好,有良好的临床应用前景。Objective To explore the feasibility and properties of a new homemade hemostatic agent prepared by the electrospun soluble eggshell membrane protein(SEP)and poly butylene succinate(PBS)with thrombin loaded.Methods The experimental hemostatic agents were prepared by the PBS/SEP.The rat hepatic hemorrhage wound model,iliac artery hemorrhage,hemostatic material implantation and oozing blood experiment at the anastomosis after incision and suturing of the dog abdominal aorta were established to evaluate the hemostatic effect and biosafety evaluation.Human embryo fibroblast cells were chosen to assessed PBS/SEP’s cytotoxicity by methyl thiazolyl tetrazolium test.Results The hemostatic time in the rat hepatic hemorrhage wound model and hemostatic effect in the iliac artery hemorrhage model of PBS/SEP was better than that of the control group,but the hemostasis time of the oozing blood experiment was slightly longer,and the difference was statistically significant(P<0.05).Cytotoxicity test show that the levels of cytotoxicity of different extracts at different time and concentrations were 0~1.In vivo experiment indicated that the implantation of rat liver might partially affected the liver function,but had no damage to the kidney function,and could not cause systemic immunity and inflammation.Conclusion PBS/SEP has a significant hemostatic effect and fine biocompatibility,which is a promising product in the clinical application in future.
