摘 要:目的观察矮地茶黄酮对肝纤维化大鼠血清肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)水平的影响,探讨矮地茶黄酮是否通过影响机体免疫功能达到防治肝纤维化的效果。方法将60只SD大鼠按随机原则分为5组(正常组,模型组,矮地茶黄酮高、中、低剂量组),每组12只。采用腹腔注射二甲基亚硝胺制备大鼠肝纤维化模型,矮地茶黄酮高、中、低剂量组通过灌服不同剂量矮地茶黄酮进行干预。采用酶联免疫吸附试验(ELISA)检测各组大鼠血清中TNF-α、IL-1β、透明质酸(HA)、层黏蛋白(LN)、Ⅳ型胶原(Ⅳ-C)、Ⅲ型前胶原(PCⅢ)水平。肝脏常规取材,病理学观察各组大鼠肝脏形态学变化,免疫组织化学方法分析波形蛋白(Vimentin)、α平滑肌肌动蛋白(α-SMA)表达情况,并进行相关性分析。结果矮地茶黄酮高、中剂量组大鼠血清中血清TNF-α、IL-1β、HA、LN、Ⅳ-C、PCⅢ水平、肝脏纤维化分级及α-SMA、Vimentin表达较模型组均明显降低,差异均有统计学意义(P<0.05)。结论矮地茶黄酮能通过降低固有免疫细胞因子TNF-α、IL-1β水平,影响机体免疫功能,进而抑制肝星状细胞增殖与活化,使细胞外基质降解增加,生成减少,进而阻止肝纤维化的发生与发展。Objective To observe the effect of the flavonoids of Ardisia japonica on serum TNF-α、IL-1β levels in hepatic fibrosis rats,and to explore the mechanism of the flavonoids of Ardisia japonica in preventing and treating hepatic fibrosis.Methods Sixty SD rats were randomly divided into 5 groups(normal group,model group,and high-dose,medium-dose and low-dose groups),12 in each group.After the model of hepatic fibrosis rats induced by dimethylnitrosamine,the flavonoids of Ardisia japonica low-dose,medium-dose and high-dose groups were intervened by different concentrations of the flavonoids of Ardisia japonica.The level of TNF-α,IL-1β,HA,LN,Ⅳ-C and PCⅢin serum of rats in each group was detected by ELISA.Liver samples were collected routinely,and morphological changes of liver were observed in each group.Immunohistochemistry was used to analyze the expression level of Vimentin andα-SMA,and the correlation was analyzed.Results Compared with the model group,the serum TNF-α,IL-1β,HA,LN,Ⅳ-C,PCⅢlevels,the grading of liver fibrosis,andα-SMA,Vimentin expression levels were significantly lower in flavonoids of Ardisia japonica high-dose,medium-dose groups(P<0.05).Conclusion The flavonoids of Ardisia japonica can affect the immune function by reducing the levels of innate immune cytokines TNF-αand IL-1β,thus inhibiting the proliferation and activation of hepatic stellate cells,increasing the degradation of extracellular matrix,and reducing the generation of extracellular matrix,thus preventing the occurrence and development of liver fibrosis.
