摘 要:目的探究白芦笋鲜汁及残渣醇提物不同极性部位对小鼠的免疫活性。方法新鲜白芦笋榨汁,残渣干燥后用15倍体积75%乙醇超声提取,共2次,每次1 h。提取物干燥所得浸膏用水分散,分别用石油醚、乙酸乙酯、正丁醇萃取,弃去水分散液中最终不溶物,浓缩干燥即得石油醚、乙酸乙酯、正丁醇、水部位提取物。通过腹腔注射80 mg·kg^(-1)环磷酰胺3 d制备免疫抑制小鼠模型。分别用白芦笋鲜汁5、10、20 mL·kg^(-1)及醇提物不同极性部位50 mg·kg^(-1)灌胃小鼠,连续7 d。观察分析小鼠的脏器指数、细胞因子含量和脾淋巴细胞增殖率,评价白芦笋鲜汁及残渣醇提物不同极性部位对小鼠免疫功能的影响。结果白芦笋鲜汁能有效地增加免疫抑制小鼠的脏器指数,提高血清中IL-2、IL-4、IFN-γ、TNF-α因子含量以及脾淋巴细胞增殖率,但各剂量组之间未发现明显的剂量-效应关系;白芦笋残渣醇提物不同极性部位中,正丁醇及水部位组小鼠的脏器指数、细胞因子含量和脾淋巴细胞增殖率明显高于模型组(P<0.05)。结论白芦笋鲜汁对免疫低下小鼠具有较好的免疫增强作用,未发现剂量-效应关系;白芦笋残渣醇提物中含有免疫活性物质,且该物质可能为高极性化合物。Objective To explore the effect of white asparagus fresh juice and different polarity fractions of ethanol extract from its residue on the immune activity in mice.Methods Fresh white asparagus was squeezed to obtain asparagus juice while the residue was dried and extracted by ultrasound with 15 times of 75%ethanol for twice(one hour each time).The obtained ethanol extract was dried to obtain powder under vacuum condition.Then the powder was suspended within water and extracted successively with petroleum ether,ethyl acetate,n-butanol followed by vacuum drying to collect petroleum ether extract,ethyl acetate extract,n-butanol extract and water extract,respectively.Immunosuppressive mice models were established by intraperitoneal injection of 80 mg·kg^(-1) cyclophosphamide for 3 days.Then white asparagus fresh juice(5,10,and 20 mL·kg^(-1))and different polarity fractions from the ethanol extract(50 mg·kg^(-1))were given to mice by gavage for 7 days.The organ index,cytokine content and splenic lymphocyte proliferation rate of the mice were analyzed,and the effect of white asparagus fresh juice and different polarity fractions of white asparagus residue on the immune function of mice was evaluated.Results White asparagus fresh juice effectively increased the organ index,the levels of IL-2,IL-4,IFN-γ,TNF-αand splenic lymphocyte proliferation rate in the immunosuppressive mice.There was no significant dose-response relationship among the groups.The organ index,cytokine content and splenic lymphocyte proliferation rate of the mice in the n-butanol and water fraction groups were significantly higher than those in the model group(P<0.05).Conclusion White asparagus fresh juice has good immunopotentiation effect on immunosuppressive mice,but no dose-response relationship is observed.The ethanol extract of white asparagus residue contains immunoactive substances,which may be high polarity compounds.
