摘  要:目的以辛伐他汀为阳性对照药,探究复方新塔花对急、慢性心肌缺血的保护作用及其初步机制。方法采用高脂乳剂+维生素D3损伤内皮建立慢性心肌缺血模型,大鼠随机分为正常组,模型组,辛伐他汀阳性药组,复方新塔花低、中、高剂量组。利用颈动脉超声观察大鼠颈动脉粥样斑块形成,评估动物模型建立,观察体质量变化,测定血脂、血栓素及相关炎症细胞因子水平、HE和油红染色观察心脏和冠状动脉组织形态。采用垂体后叶素(Pit)诱导急性心肌缺血大鼠模型,大鼠随机分为正常组,模型组,辛伐他汀阳性药组,复方新塔花低、中、高剂量组。观察心电图ST段变化、血清肌酸激酶(CK-MB)、乳酸脱氢酶(LDHA)和肌钙蛋白(cTn1)的水平及心脏组织病理。结果复方新塔花3个剂量组均可降低血脂、IL-1β、TNF-α、IL-6的水平以及血栓素的含量,改善B超显示的血管内壁增厚、脊柱斑块等表现;改善内膜增厚,炎性细胞浸润,斑块形成等病理变化,改善主动脉内、中、外等主动脉膜各层的脂质沉积,钙化等油红染色变化。复方新塔花3个剂量组可对抗Pit引起的急性心肌缺血ST段偏移,降低血清CK-MB、LDHA水平及cTn1含量;改善心肌缺血引起的心肌纤维排列紊乱,炎性细胞浸润等病理变化。结论复方新塔花对急慢性心肌缺血具有显著的保护作用。Objective To determine the protective effect of compound Xinta flower on acute and chronic myocardial ischemia.Methods The chronic myocardial ischemia model was established with high-fat emulsion+vitamin D3 to damage the endothelium.Rats were randomly divided into a normal group,a model group,a simvastatin-positive drug group,and compound Xinta flower low,medium,and high dose groups.The formation of carotid atherosclerotic plaque,establishment of rat models,change in the body mass,and level of blood lipids,thromboxane and related inflammatory cytokines were observed with carotid artery ultrasound.The heart and coronary artery tissue morphology was observed by HE and oil red staining.The rat model of acute myocardial ischemia was induced by pituitrin,and the experimental rats were divided into 6 groups.The ST segment changes in electrocardiogram,serum creatine kinase(CK-MB),lactate dehydrogenase(LDHA)and troponin(cTn1)levels and cardiac tissue pathology were observed.Results The low,medium,and high dose compound Xinta flower all reduced the level of blood lipids,IL-1β,TNF-α,IL-6,and the content of thromboxane,and improved the performance of B-ultrasound,such as the thickening of the inner wall of blood vessels and spinal plaque.Intimal thickening,inflammatory cell infiltration,plaque formation and other pathological changes in HE staining,and aortic membrane lipid deposition,calcification and other oil red staining changes in the aortic membrane were improved.Compound Xinta flower resisted the ST segment deviation caused by pituitary vasopressin,reduced the serum CK-MB,LDHA levels and cTn1 content,and improved myocardial fiber arrangement disorder caused by myocardial ischemia,inflammation pathological changes such as cell infiltration.Conclusion Compound Xinta flower can protect against acute and chronic myocardial ischemia.
