摘　　要：目的筛选枳壳抗结肠癌药效物质组分的最佳提取工艺。方法采用正交设计,对影响提取工艺的3个因素(溶剂用量、提取时间、提取温度)进行考察,以人结肠癌细胞系HT-29细胞抑制率为考察指标,筛选枳壳抗结肠癌药效物质组分的最佳提取工艺。同时,结合层次分析法,筛选以黄酮含量和出膏率为综合评价指标的权重系数及其范围,并采用Pearson相关性分析对结果进行验证。结果枳壳黄酮最佳提取工艺为25倍量75%乙醇,在75℃水浴条件下,回流提取2.5 h。以黄酮含量和出膏率为指标的综合权重系数范围为0.75~0.90、0.10~0.25。结论该方法在保证了枳壳治疗结肠肿瘤药效的基础上,简化了考察指标,为枳壳的深入利用和开发提供了实验基础。Objective To optimize the extraction of active components from Fructus aurantii.Methods Three factors(solvent dosage,extraction time and extraction temperature)affecting the extraction process were investigated by orthogonal design.The inhibition rate of human colon cancer cell line HT-29 was used as the index to screen the optimal extraction of the active components of Fructus aurantii.At the same time,combining analytic hierarchy process,the weight coefficient and the range of total flavonoids content,and the extract in rate were chosen as comprehensive evaluation indexes.Pearson correlation analysis was used to verify the results.Results The optimum extraction of flavonoids from Fructus aurantii was 25 times of 75%ethanol,refluxing extraction for 2.5 h at 75℃.The comprehensive weight coefficient ranged from 0.75 to 0.90 for total flavonoids content and from 0.10 to 0.25 for the extract in rate.Conclusion The method ensures the efficacy of Fructus aurantii for colon cancer,simplifies the evaluation index,and provides experimental basis for the further utilization and development of Fructus aurantii.
