摘  要:目的:探讨苗药抑癌汤浓缩液对人肺癌NCI-H446细胞的影响。方法:培养细胞加不同剂量苗药抑癌汤药处理,用CCK-8及流式检测细胞凋亡筛选苗药抑癌汤最佳浓度。按照最佳浓度处理细胞将细胞分为抑癌汤组(2.5 g/mL)顺铂组(0.6μg/mL)、顺铂(0.6μg/mL)+二十分之一(0.125 g/mL)抑癌汤组、空白对照组,流式细胞术检测细胞周期、凋亡,划痕实验检测细胞迁移能力,Transwell实验检测细胞侵袭能力。结果:与对照组比较其余三组细胞活力显著下降(P<0.05),细胞凋亡率提高,细胞周期停滞于G2期。结论:抑癌汤能显著抑制NCI-H446细胞增殖、迁移与侵袭,并诱导NCI-H446细胞凋亡,且抑癌汤结合顺铂效果更佳。Objective:To explore the effect of concentrate solution of Miao medicine Yi-Ai-Tang(YAT)on human lung cancer NCI-H446 cells.Methods:Treatment of cultured cells with different doses of YAT.The best concentration of YAT was determined by using CCK-8 and flow cytometry to detect apoptosis.According to the optimal concentration,the cells were divided into four groups:YAT group,cisplatin group(0.6μg/mL),cisplatin group(0.6μg/mL)plus one twentieth(0.125 g/mL)YAT group and blank control group.Cell cycle and apoptosis were detected by flow cytometry,scratch test and transwell test were used to detect cell migration and invasion respectively.Results:Compared with the control group,the cell viability of the other three groups decreased significantly(P<0.05),the apoptosis rate increased and cell cycle arrest in G2 phase.Conclusion:YAT significantly inhibit the proliferation,migration and invasion of NCI-H446 cells,and induce the apoptosis of NCI-H446 cells,the combination of YAT and cisplatin has better effect.
