碱性嗜热SGNH家族酯酶的异源功能研究

摘  要:微生物来源的SGNH家族水解酶,作为一种嗜热菌,其菌体内嗜热酶具有较高稳定性与耐高温性,在较高温度环境下表现出良好的催化活性,具有较大的工业应用价值。水解酶序列区内4个催化位点分别是Ser,Gly,Asn,His,也因此称为SGNH家族水解酶。我们通过SGNH家族水解酶催化反应,质子变换位置产生激活效应促进底物与酶进行催化反应,从而形成四面形状的复合物,利用催化反应原理在工业中合成β-内酰胺类抗生素主要原料。在重组酶实验中具体对照温度以及时间,进行金属离子反应活性测试,测试结果表明较低长度的酰基链底物反应活性较好,因此,进一步确认SGNH家族酯酶的酶学性质较好,有利于应用在主要β-内酰胺类抗生素工业制备过程。As a kind of thermophilic bacteria,the hydrolase of the SGNH family derived from microorganisms has high stability and high temperature resistance,and exhibits good catalytic activity under high temperature environment,which has great industrial application value.The four catalytic sites in the hydrolase sequence are respectively Ser,Gly,Asn,His,and are also called SGNH family hydrolases for this reason.Through the catalyzed reaction by SGNH family hydrolase,and the proton position conversion produces activation effect to promote the substrate to catalyze the reaction with the enzyme,thus forming a four-shaped complex.By using the catalytic reaction principle,β-lactam antibiotics in the industrial synthesis are made as the main raw materials.In recombinant enzyme experiments,compared with the experimental temperature and time,metal ion reactivity test has been made,the test shows that the lower the length of the acyl chains the substrate has good reactivity.Therefore,it further confirms that the property of SGNH family esterase is good,is advantageous to the main application inβ-lactam antibiotics preparation of industrial process.
