摘 要:CONSTANS(CO)基因是植物光周期途径中重要的转录调控因子。为了探究甘蓝型油菜CO同源基因的功能,以拟南芥CO基因的CDS序列为参考序列,从甘蓝型油菜品种Westar的cDNA中分离得到CO的两个同源基因(基因编号:BnaC09g41990D和BnaA10g18430D),命名为BnaC09CO和BnaA10CO。构建双35S超表达载体转化哥伦比亚拟南芥,得到超表达转基因株系,观察表型并进行定量分析。结果表明超表达转基因株系的开花时间比野生型早3~5 d,BnaC09CO、BnaA10CO、FT与SOC1表达量较野生型显著升高。说明在长日照条件下甘蓝型油菜BnaC09CO和BnaA10CO基因起到了与CO基因类似的作用,能够引起拟南芥FT和SOC1的上调表达,从而促进拟南芥的开花。CONSTANS(CO) gene is an important transcriptional regulator in photoperiodic flowering pathway.To explore the function of CO homologous gene of Brassica napus, CDS sequence of Arabidopsis CO was used as reference, 2 homologs(BnaC09 g41990 D, BnaA10 g18430 D) were isolated from cDNA of Brassica napus variety Westar and named BnaC09 CO and BnaA10 CO respectively. The overexpression vectors were constructed to transform into Arabidopsis thaliana to observe phenotype and quantitative analysis. Results showed that flowering time of overexpression lines was 3-5 days earlier than that of wild type, and the expression of BnaC09 CO, BnaA10 CO, FT and SOC1 were significantly higher than those of wild type. These indicated that BnaC09 CO and BnaA10 CO played similar roles to CO, which could upregulate the expression of FT and SOC1 in Arabidopsis thaliana to promote flowering under long-day condition.
