摘 要:目的:研究运动疲劳对大鼠焦虑情绪的影响,观测海马CA1区场电位θ节律震荡的变化,揭示运动疲劳引发不良情绪效应的可能神经电生理机制。方法:雄性Wistar大鼠随机分为对照组(control)和运动疲劳组(exercise training,ET),采用反复7 d力竭跑台训练方式建立疲劳模型。行为学实验采用高架十字迷宫观察大鼠焦虑行为的改变。随后,利用在体电生理记录方法,比较两组大鼠海马局部场电位θ节律活动的差异。结果:高架十字迷宫检测发现,ET组大鼠进入高架十字迷宫开放臂次数较对照组大鼠明显减少(P<0.05),并且在其中停留的时间也显著性缩短(P<0.01);同时,在闭合臂的活动时间大幅延长(P<0.01)。在体海马CA1区θ节律电信号记录显示,ET组大鼠功率峰值显著高于对照组大鼠(P<0.05),而功率峰值对应的θ节律频率值不存在组间差异(P>0.05)。结论:运动疲劳后大鼠探索行为减少,焦虑样行为增多,并伴有海马CA1区θ节律活动增强。这种焦虑情绪与θ节律震荡活动的改变相关,提示θ节律场电位信号活动的增强,可能是运动疲劳导致焦虑等不良情绪产生的电生理机制之一。Objective:To study the effect of exercise fatigue on the anxiety of rats,observe the changes of theta rhythm in hippocampal CA1 region,and reveal the possible neuro-electrophysiological mechanism of exercise fatigue inducing adverse emotional effects.Methods:Male Wistar rats were randomly divided into control group(control)and exercise training group(ET).The fatigue model was established by repeated 7 dayst’readmill exhausted exercise.Elevated plus maze test was employed to detect the rats’anxiety-like behaviors after the exercise modeling.Subsequently,CA1 region multi-channel local field potentials(LFP)recording in vivo.Comparing the difference of theta rhythm activity in hippocampus between two groups of rats.Results:In elevated plus maze test,compared with control group,the entry times to the open arm was significantly decreased compared with ET group(P<0.05),so as the time of rats staying in the open arm(P<0.01);And the time ET group staying in closed arm was significantly longer than control group(P<0.01).The electrical signal record of theta rhythm in the hippocampal CA1 region showed that the peak power of the ET group was significantly higher than that of the control group(P<0.05).However,we did not find any significant difference between two groups on theta frequency(P>0.05).Conclusion:After exercise fatigue,rats’exploratory behaviors decrease,anxiety-like behaviors increase,and theta rhythm in hippocampal CA1 region increases.This kind of anxiety is related to the change of theta rhythm,suggesting that the enhancement of theta rhythm may be one of the electrophysiological mechanisms of anxiety and other bad emotions caused by exercise fatigue.
