摘 要:目的:建立利肝宁片中对羟基苯乙酮和绿原酸的含量测定方法,为利肝宁片的质量控制提供依据。方法:采用紫外双波长同时测定的反相高效液相色谱法。色谱柱:安捷伦ZORBAX Eclipse XDB C18;流动相:(体积分数)0.05%磷酸溶液-乙腈(9010);流速:1.0 mL·min^(-1);柱温:35℃;检测波长:275 nm(对羟基苯乙酮)和327nm(绿原酸)。结果:在选定的色谱条件下,对羟基苯乙酮峰和绿原酸峰与其他峰分离度良好。对羟基苯乙酮浓度在2.545~20.36 mg·L^(-1)范围内与峰面积有良好的线性关系(r=0.9997);平均回收率(n=6)为98.97%,RSD=1.72%。绿原酸浓度在10.31~82.47 mg·L^(-1)范围内,与峰面积有良好的线性关系(r=0.9998);平均回收率(n=6)为98.05%,RSD=1.04%。结论:该方法专属性强、灵敏度高,可用于利肝宁片中对羟基苯乙酮和绿原酸的含量测定。Objective:To establish a content determination method for hydroxyacetophenone and chlorogenic acid in Liganning Tablets and provide the basis of the quality control.Methods:RP-HPLC measurement using two different UV wavelengths simultaneously was adopted.Column ZORBAX Eclipse XDB C18,mobile phase was Phosphoric acid 0.05%Acetonitrile(90:10),the flow rate was 1.0 mL·min^(-1),the column temperature 35℃,the detection wavelength was 275 nm for hydroxyacetophenone and 327 nm for chlorogenic acid.Results:The hydroxyacetophenone and chlorogenic acid can completely separate from the other component in the selected chromatographic conditions.Of hydroxyacetophenone:Good linear relationship was presented in the range of 2.545-20.36 mg·L^(-1)(r=0.9997).The average recovery rate was 98.97%,RSD=1.72%,(n=6).Of chlorogenic acid:A good linear relationship was found in the range of 10.31-82.47 mg·L^(-1)(r=0.9997).The average recovery was 98.05%,RSD=1.04%(n=6).Conclusion:The method is specific,sensitive,and can be used in the content determination of hydroxyacetophenone and chlorogenic acid content in Liganning tablets.
