摘  要:目的:基于网络药理学探讨赤芍治疗黄疸的作用机制。方法:利用TSMSP数据库筛选赤芍的有效成分。多个数据库联合挖掘药物预测靶点和黄疸的治疗靶点,分析两个靶点集合并找出交集靶点。通过Cytoscape绘制药物有效成分-潜在治疗靶点网络图,采用String数据库构建蛋白互作网络,Cytoscape软件MCODE插件进行模块化分析,并运用R软件及clusterProfiler对交集靶点进行GO和KEGG富集分析。结果:筛选出赤芍14个有效成分及对应靶点228个,黄疸相关靶点2155个,交集靶点59个。PPI网络共包含55个节点,187条连接,靶点NR1H4处于核心位置。这些靶点主要涉及脂质定位、类固醇代谢过程、脂质运输等生物学过程,主要参与了胆汁分泌、ATP结合盒式转运蛋白、PPAR信号通路等生物通路。结论:赤芍主要通过调节胆汁分泌和转运、提高药物代谢酶含量等方面治疗黄疸,运用赤芍治疗胆汁淤积性黄疸或药物性肝损伤、炎症引起的肝细胞性黄疸可能取得较好疗效。Objective:To investigate the mechanism of Radix Paeoniae Rubra(RPR)in jaundice treatment based on Network Pharmacology.Methods:The effective components of RPR were obtained by the TCMSP database.Multiple databases were used to screen potential drug targets and therapeutic targets for jaundice,and cross-analyzing the two sets of targets to establish intersection targets.Then using Cytoscape to visualization the effective drug components-potential therapeutic targets network.And the protein-protein interaction(PPI)network was constructed by the String database.The plug-in MCODE was used for modular analysis.R and clusterProfiler were used in the Functional Enrichment Analysis of intersection targets of GO and KEGG.Results:In this investigation,14 effective components with 228 corresponding targets in RPR,2155 jaundice-related targets,and 59 intersection targets were screened.55 nodes and 187 connections consisted in the PPI network and the target NR1 H4 is central to it.Biological processes such as lipid localization,steroid metabolism,lipid transport,phosphate transport were related to these targets,which are mainly involved in bile secretion,ABC transporters,and PPAR signaling pathway,etc.Conclusion:PRP mainly by increasing the secretion and transport of bile,improving the quantity of drug metabolic enzymes in jaundice treatment.It may have a better effect by using PRP to treat cholestatic jaundice or hepatocellular jaundice caused by drug-induced liver injury or inflammation.
