摘 要:目的:利用脂质组学的方法探讨"上火"发生的机制。方法:利用多维质谱"鸟枪法"脂质组学技术(MDMS-SL)完成了93例各类"上火"患者以及37名正常人群血浆中多种脂质分子的定性定量分析,其中"上火"患者中36例以口腔溃疡为主要表现,32例以干眼为主要表现,25例以牙龈炎主要表现。结果:与正常人群比较,"上火"人群血浆中缩醛型磷酯、sn-2型溶血磷酯、神经酰胺、磷酯酰肌醇(PI)发生了显著变化,其中以PI变化最为明显,具体为"上火"人群PI总体含量比正常人群上升约44%(P<0.05)。不同"上火"表型的人群在不同脂质的代谢方面表现出相同的变化趋势。结论:"上火"人群的血浆脂质代谢发生了显著变化,脂质代谢的异常可能通过氧化应激以及能量代谢增强,导致"上火"症状的发生。Objective: To explore the mechanism of ’shanghuo’ by the lipidomics. Methods: The qualitative and quantitative analysis of various lipid molecules in plasma about 93 ’shanghuo’ patients(who includes 36 mouth ulcer, 32 xerophthalmia and 25 gingivitis) and 37 healthy controls was completed by using multi-dimensional mass spectrometry ’shotgun’ lipidomics(MDMS-SL). Results: Compared with the healthy people, the content of acetal phosphatidylethanolamine, sn-2 position lysophosphatidylethanolamine, ceramide and phosphatidylinositol in the ’shanghuo’ changed significantly, among which the PI was the most obvious. The overall content of PI in the ’shanghuo’ people was about 44% higher than that in the healthy controls(P<0.05). People with different ’shanghuo’ symptoms showed the same trend in different lipid metabolism. Conclusion: There are significant changes in plasma lipid metabolism in ’shanghuo’ paitents. The disorder of lipid metabolism may lead to the occurrence of ’shanghuo’ symptoms through the oxidative stress and energy metabolism enhancement.
