摘 要:目的:探讨艾条悬灸对皮质酮肾阳虚大鼠海马及杏仁核中盐皮质激素受体(MR)和糖皮质激素受体(GR)基因及蛋白表达的影响,揭示艾条悬灸改善肾阳虚证的可能机制。方法:70只SD雄性大鼠随机分为空白对照组、造模组。造模组大鼠给予氢化可的松注射液肌注14d建立肾阳虚模型,造模成功的40只大鼠再随机分成模型组、艾条悬灸组。艾条悬灸组予以艾条悬灸关元、肾俞穴(双),每次20min,每日1次,共14次。采用ELISA方法检测大鼠血清CRH、ACTH、CORT含量,采用RT-PCR和Western blot方法检测各组大鼠海马和杏仁核中MR和GR mRNA及蛋白的表达。结果:与空白对照组比较,模型组大鼠血清CRH、ACTH、CORT含量显著下降(P<0.01);经艾灸治疗后,大鼠血清CRH、ACTH、CORT含量较模型组均显著上升(P<0.05,P<0.01)。与空白对照组比较,模型组海马GR mRNA和蛋白表达显著下降(P<0.05);经艾灸治疗后,海马GR mRNA和蛋白表达显著增加(P<0.05,P<0.01)。与空白对照组比较,模型组杏仁核MR、GR mRNA和蛋白表达显著下降(P<0.05,P<0.01);经艾灸治疗后,杏仁核MR、GR mRNA和蛋白表达显著增加(P<0.05,P<0.01)。结论:艾条悬灸能升高肾阳虚大鼠血清ACTH、CRH、CORT的水平;能上调海马、杏仁核GR mRNA及蛋白表达,上调杏仁核的MR mRNA及蛋白表达,这可能是艾灸改善肾阳虚证的分子机制之一。Objective: To investigate the effects of moxibustion on the expressions of mineralocorticoid receptor(MR) and glucocorticoid receptor(GR) gene and protein in hippocampus and amygdala of rats with corticosterone kidneyyang deficiency, and to reveal the possible molecular mechanism of moxibustion improving kidney-yang deficiency syndrome. Methods: A total of 70 SD male rats were randomly divided into blank control group and modeling group. Rats in the modeling group were given Hydrocortisone Intramuscular Injection for 14 days to establish the kidney-yang deficiency rat model. Forty rats successfully modeled were randomly divided into the model group and the moxibustion group. In the moxibustion group, suspended moxibustion with moxa sticks was applied to Guanyuan(RV5) and Shenshu(BL25), 20 min at a time, once a day, which was treated for a total of 14 times. After the treatment, the contents of CRH, ACTH and CORT in serum of rats were detected by ELISA, and the mRNA and protein expressions of MR and GR in hippocampus and amygdala in each group rats were detected by RT-PCR and Western blot. Results: Compared with the blank control group, the serum contents of CRH, ACTH and CORT in the model group rats were significantly decreased(P<0.01). After treating with moxibustion, the serum contents of CRH, ACTH and CORT in rats were increased significantly(P<0.05, P<0.01). Compared with the blank control group, the mRNA and protein expression of GR in hippocampus in the model group rats was decreased significantly(P<0.05). After treating with moxibustion, the mRNA and protein expression of GR in hippocampal was significantly increased(P<0.05, P<0.01). Compared with the blank control group, the mRNA and protein expressions of MR and GR in the amygdala in the model group were significantly decreased(P<0.05, P<0.01). After treating moxibustion, the mRNA and protein expressions of MR and GR in amygdala were significantly increased(P<0.05, P<0.01).
