摘  要:基于超高液相色谱-串联四极杆/线性离子阱质谱(QTRAP UPLC-MS/MS),建立了尿液中30种滥用药物的筛查方法。采用蛋白沉淀法处理尿液样品,实现对多类别滥用药物的高效提取。采用分段多反应监测(s MRM)联合信息依赖性采集(IDA)与增强离子扫描(EPI)模式,结合EPI谱库检索匹配确证检出物信息,并引入内标辅助定量。30种滥用药物质量浓度在0.5~50 ng/mL范围内线性关系良好(R^(2)> 0.99);检出限为0.01~0.25 ng/mL,定量限为0.1~0.4 ng/mL;加标回收率为76.2%~112.5%,相对标准偏差为3.1%~12%。该方法适用于实际尿样中痕量滥用药物的定性与定量分析。A systematic screening method for 30 abused drugs in urine was developed using ultra-performance liquid chromatography-tandem hybrid triple-quadrupole linear ion trap mass spectrometry(QTRAP UPLC-MS/MS).The protein precipitation method was used for sample pretreatment to achieve efficient extractions of different types of abused drugs.Scheduled multiple reaction monitoring(s MRM) combined with informationdependent acquisition(IDA) and enhanced ion(EPI) scanning mode was applied in data acquisition.The information of detected compounds could be confirmed by EPI mass spectral library research,and the internal standard was introduced for quantitative analysis.30 abused drugs showed good linear relationships in the concentration range of 0.5-50 ng/m L with R^(2)> 0.99.Limits of detection were 0.01-0.25 ng/m L,while limits of quantitation were 0.1-0.4 ng/m L.Spiked recoveries at low,medium,and high concentrations were76.2%-112.5%,with relative standard deviations of 3.1%-12%.This method is suitable for qualitative and quantitative analysis of trace abused drugs in urine samples.
