摘 要:目的:探讨芍药苷对含糖透析液作用下人腹膜间皮细胞的α-SMA和CTGF的影响。方法:使用永生人HMrSV5细胞株,分为空白组、高糖组、芍药苷低剂量组、芍药苷中剂量组、芍药苷高剂量组,观察各组在4.25%含糖透析液作用下α-SMA和CTGF的变化。Western Blot检测各组α-SMA和CTGF的表达,免疫荧光法观察α-SMA表达的变化。其中芍药苷低剂量组浓度为50μg/mL、芍药苷中剂量组浓度为100μg/mL、芍药苷高剂量组浓度为200μg/mL。结果:高糖组的α-SMA的表达高于空白组、芍药苷低剂量组、芍药苷中剂量组和芍药苷高剂量组(P<0.05)。芍药苷高剂量组α-SMA的表达低于芍药苷中剂量组和芍药苷低剂量(P<0.05)。芍药苷高剂量组各组间随浓度增加α-SMA的表达逐渐减少(P<0.05)。间接免疫荧光显示高糖组α-SMA的表达明显增强,芍药苷组随浓度增加α-SMA的表达逐渐减少。高糖组CTGF的表达与空白组、芍药苷高剂量组有显著差异(P<0.05),与芍药苷中剂量组和芍药苷低剂量无显著差异(P>0.05)。结论:芍药苷可以下调高糖作用下腹膜间皮α-SMA和CTGF的表达,可能减缓腹膜纤维化的进展。Objective:To explore the effect of paeoniflorin onα-SMA and CTGF of human peritoneal mesothelial cells under the action of sugar-containing dialysate.Methods:Using immortalized human HMrSV5 cell line,it was divided into blank group,high glucose group,low-dose group,paeoniflorin middle-dose group,paeoniflorin high-dose group,and observed each group under the action of 4.25%sugary dialysate-Changes in SMA and CTGF.Western Blot detected the expression ofα-SMA and CTGF in each group,and detectedα-SMA by immunofluorescence.The concentration of the paeoniflorin low-dose group was 50μg/mL,the paeoniflorin medium-dose group concentration was 100μg/mL,and the paeoniflorin high-dose group concentration was 200μg/mL.Results:The expression ofα-SMA in the high glucose group was higher than that in the blank group,paeoniflorin low-dose group,paeoniflorin middle-dose group and paeoniflorin high-dose group(P<0.05).The expression ofα-SMA in the paeoniflorin high-dose group was lower than that in the paeoniflorin middle-dose group and the paeoniflorin low-dose group(P<0.05).The expression ofα-SMA gradually decreased with increasing concentration in each group of paeoniflorin high-dose group(P<0.05).Indirect immunofluorescence showed that the expression ofα-SMA in the high glucose group was significantly enhanced,and the expression ofα-SMA in the paeoniflorin group gradually decreased with increasing concentration.The expression of CTGF in the high glucose group was significantly different from that in the blank group and the high-dose paeoniflorin group(P<0.05),and was not significantly different from that in the middle-dose paeoniflorin group and the low-dose paeoniflorin group(P>0.05).Conclusion:Paeoniflorin can down-regulate the expression ofα-SMA and CTGF in peritoneal mesothelium under the action of high glucose,which may slow the progress of peritoneal fibrosis.
