摘  要:【目的】了解我国华南沿海黄鳍棘鲷(Acanthopagrus latus)种群的遗传差异和分化情况,为促进华南沿海黄鳍棘鲷资源的可持续利用及遗传多样性保护提供参考依据。【方法】采用基于线粒体细胞色素b基因(Cytb)序列的分析方法,对福建厦门(XM)、广东汕尾(SW)、广东阳江(YJ)、海南海口(HK)、海南三亚(SY)、广西北海(BH)、广西钦州(QZ)和广西防城港东兴(FC)等8个黄鳍棘鲷种群进行遗传结构分析。【结果】经PCR扩增和测序获得8个黄鳍棘鲷群体320条Cytb基因的部分序列(1141 bp),其中碱基A+T平均含量(55.1%)高于碱基C+G平均含量(44.9%),表现出较强的少G偏倚性,编码380个氨基酸残基,定义62个单倍型。总的单倍型多样性指数(Hd)和核苷酸多样性指数(π)分别为0.78638和0.00164;群体间遗传距离、遗传分化系数(Fst)和基因流(Nm)分别为0.00093~0.00232、-0.01300~0.56149和0.39049~inf。非加权组平均法(UPGMA)系统发育进化树分析结果表明,大部分单倍型以琼州海峡为界可划分为东海域(EAST)和西海域(WEST)两个组群。聚类分析结果也表明8个黄鳍棘鲷群体以琼州海峡为界分隔为两类。分子方差分析(AMOVA)结果显示,组群间、群体内个体间的遗传变异分别为41.26%和58.65%。Tajima’s D和Fu’s Fs检验及核苷酸错配分析结果均表明,华南沿海黄鳍棘鲷可能经历了种群扩张事件。【结论】华南沿海黄鳍棘鲷以琼州海峡为界分隔的东海域和西海域两个组群间的遗传分化明显,可将其作为2个管理单位(MUs)进行渔业管理。【Objective】Genetic structure analysis of Acanthopagrus latus population was studied to provide reference for promoting the sustainable use and the protection of genetic diversity of A.latus resources along the South China coast.【Method】The mitochondrial b gene(Cytb)sequence analysis method was used to analyze the genetic structure of eight A.latus populations from Xiamen,Fujian(XM),Shanwei,Guangdong(SW),Yangjiang,Guangdong(YJ),Haikou,Hainan(HK),Sanya,Hainan(SY),and Beihai,Guangxi(BH),Qinzhou,Guangxi(QZ),and Dongxing,Fangcheng Harbor,Guangxi(FC).【Result】PCR amplification and sequencing obtained 320 Cytb gene partial sequences(1141 bp)in 8 A.latus populations,of which the average content of A+T(55.1%)was higher than C+G(44.9%),showing a strong less G Bias.The sequence encoded 380 amino acids and defined 62 haplotypes.The total haplotype diversity index(Hd)and nucleotide diversity index(π)were 0.78638 and 0.00164,respectively.The genetic distance,genetic differentiation coefficient(Fst)and gene flow between the populations were 0.00093-0.00232,-0.01300-0.56149 and 0.39049-inf,respectively.The Analysis results of UPGMA relationship tree indicated that most of the haplotypes were divided into two groups of East Sea(EAST)and West Sea(WEST)with the Qiongzhou strait as the boundary.Cluster analysis results also showed that the eight populations were separated into two groups by the Qiongzhou Strait.Molecular variance(AMOVA)analysis results showed that the genetic variation between groups and individuals within the groups was 41.26%and 58.65%,respectively.Tajima’s D and Fu’s Fs tests results,as well as nucleotide mismatch analysis,indicated that A.latus along the South China coast might have experienced a population expansion event.【Conclusion】Genetic differentiation between the A.latus in the East Sea and West Sea separated by Qiongzhou Strait are significant.Therefore,the A.latus along the South China coast can be used as two management units for fishery.
