摘  要:为揭示亚洲小车蝗Oedaleus asiaticus两型现象转变的分子机制,本研究以亚洲小车蝗3龄蝗蝻为研究对象,对散居型单头饲养的亚洲小车蝗进行高密度群居化胁迫处理,并对处理后型变第1、3、5、7天的蝗蝻取样,利用Illummina Hiseq 4000高通量测序平台对样本转录组进行测序、组装、注释,筛选调控亚洲小车蝗由散居型向群居型转变的相关基因。测序后共获得60502个unigenes,在Nr、KEGG、KOG、SwissPort四大数据库中比对得到有注释信息基因数目分别为27820、21529、14346和15154个,型变过程中差异表达基因数目在第3天时最多,为7478个,第7天时差异表达基因数目最少,为1857个;亚洲小车蝗3龄蝗蝻型变过程差异表达基因的GO功能主要富集于生物过程、细胞组分和分子功能3个部分,高密度环境诱导下亚洲小车蝗3龄蝗蝻型变第1、3、5、7天差异表达基因的数量呈现先增加后减少的趋势,型变第1天时556个差异表达基因显著富集到120条代谢通路上,型变第3天时1133个差异表达基因显著富集到130条代谢通路上,型变第5天时374个差异表达基因显著富集到115条代谢通路上,型变第7天时220个差异表达基因显著富集到98条代谢通路上。In order to reveal the molecular mechanism of the phenotypic transition of grasshopper Oedaleus asiaticus,the 3 rd instars of O.asiaticus were treated with high-density swarming stress in this study.The transcriptome was sequenced,assembled,and annotated using the Illummina Hiseq 4000 high-throughput sequencing platform to screen for genes regulating the shift from dispersal to swarming in O.asiaticus.A total of 60502 unigenes were obtained after sequencing,and the numbers of annotated genes were 27820,21529,14346,and 15154 in the four major databases of Nr,KEGG,KOG,and SwissPort.The numbers of differentially expressed genes were 7478 on day 3 and 1857 on day 7,and the GO functions of differentially expressed genes were mainly enriched in three parts:biological processes,cellular components,and molecular functions.On day 1,556 differentially expressed genes were significantly enriched to 120 metabolic pathways;on day 3,1133 differentially expressed genes were significantly enriched to 130 metabolic pathways;on day 5,374 differentially expressed genes were significantly enriched to 115 metabolic pathways;on day 7,220 differentially expressed genes were significantly enriched to 98 metabolic pathways.
