摘  要:目的对溪黄草三种基源植物的成份进行比较,为评价溪黄草药材的质量提供理论依据。方法采用Boston Green ODS C 18色谱柱(200×4.6 mm,5μm,Boston Green ODS),以乙腈-0.1%磷酸水(25∶75)为流动相,等梯度洗脱,流速为1 mL·min^(-1),检测波长为330 nm,柱温为25℃,测定迷迭香素和咖啡酸的含量;以甲醇-0.1%磷酸水(90∶10)为流动相,等梯度洗脱,流速为1 mL·min^(-1),检测波长为210 nm,柱温25℃,测定熊果酸和科罗索酸的含量。结果HPLC方法简便,专属性强。迷迭香酸在0.010 mg·mL^(-1)~0.060 mg·mL^(-1)(r=0.9995)范围内线性良好;平均加样回收率为98.9%,RSD值为2.31%。咖啡酸在0.010 mg·mL^(-1)~0.09 mg·mL^(-1)(r=0.9996)范围内线性良好;咖啡酸的平均加样回收率为97.2%,RSD为1.91%。科罗索酸在0.037 mg·mL^(-1)~0.294 mg·mL^(-1)(r=0.9994)范围内线性良好;科罗索酸的平均加样回收率为95.2%,RSD值为1.91%。熊果酸在0.057 mg·mL^(-1)~0.453 mg·mL^(-1)(r=0.9991)范围内线性良好;熊果酸的平均加样回收率为100.4%,RSD值为2.64%。结论溪黄草不同基源植物的成分含量有显著差异,溪黄草的质量评价标准有待进一步深入研究。OBJECTIVE Comparing the components of three basic plants of Isodon serra,it provides a theoretical basis for evaluating the quality of Isodon serra herbal materials.METHODS The Boston Green ODS C 18 column(200×4.6 mm,5μm,Boston Green ODS)was used as the mobile phase with acetonitrile-0.1%phosphoric acid water(25∶75).The gradient was eluted at a flow rate of 1 mL·min^(-1).The wavelength was 330 nm,the column temperature was 25℃,and the content of rosmarinin and caffeic acid was determined.The mobile phase was methanol-0.1%phosphoric acid(90∶10),and the gradient was eluted at a flow rate of 1 mL·min^(-1).The content of ursolic acid and corosolic acid was determined at 210 nm and a column temperature of 25°.RESULTS The HPLC method is simple and specific.The rosmarinic acid showed good linearity in the range of 0.10 mg·mL^(-1)to 0.60 mg·mL^(-1)(r=0.9995);the average recovery of the sample was 98.9%,and the RSD value was 2.31%.The caffeic acid showed good linearity in the range of 0.010 mg·mL^(-1)to 0.09 mg·mL^(-1)(r=0.9996);the average recovery of caffeic acid was 97.2%,and the RSD was 1.91%.Corosolic acid showed good linearity in the range of 0.037 mg·mL^(-1)to 0.294 mg·mL^(-1)(r=0.9994);the average recovery of corosolic acid was 95.2%,and the RSD value was 1.91%.Ursolic acid was linearly in the range of 0.057 mg·mL^(-1)to 0.453 mg·mL^(-1)(r=0.9991);the average recovery of ursolic acid was 100.4%,and the RSD value was 2.64%.CONCLUSION There are significant differences in the content of different base plants in Isodon serra.The quality evaluation criteria of Isodon serra needs further study.
