中药复方合剂抗甲型H1N1流感病毒活性的实验研究

摘　　要：目的:观察中药复方合剂对甲型流感病毒A/PR/8/34(H1N1)的抑制作用。方法:采用鸡胚法检测药物对鸡胚的最大无毒限量和半数鸡胚感染量(EID50),通过中药复方合剂对病毒的直接作用、预防作用和治疗作用,观察鸡胚的存活率和测定鸡胚尿囊液血凝滴度。结果:中药复方合剂对鸡胚最大安全浓度为500 mg/mL,半数鸡胚感染量(EID50)为10-3.83;在3种给药方式中,中药复方合剂各剂量组对鸡胚均有不同程度的保护作用,250mg/mL时鸡胚存活率100%;在血凝滴度测试中,同时给药、预防给药和治疗给药药物浓度在62.5、125.0、250.0 mg/mL时,血凝滴度均低于病毒对照组的1/4,表现为抗病毒活性(P<0.01),且呈现一定的量效关系。结论:中药复方合剂3种给药方式均能明显抑制甲型流感病毒A/PR/8/34(H1N1)在鸡胚中的增殖,对感染鸡胚有较好的保护作用。Objective:To observe the inhibitory effect of Chinese Herbal Compound Mixture on influenza A virus A/PR/8/34(H1N1).Methods:Embryonated chicken eggs were used to detect the maximum non-toxic limit of the drug and half of the chicken embryo infection dose EID50.Observe the survival rate of chicken embryos and determine the hemagglutination titer of chicken embryo allantoic fluid through the direct,preventive and therapeutic effects of Chinese Herbal Compound Mixture on the virus.Results:The maximum safe concentration of Chinese Herbal Compound Mixture on chicken embryos was 500 mg/mL,and the EID50 for half of chicken embryos was 10-3.83.Among the three modes of administration,each dose group of Chinese Herbal Compound Mixture has different degrees of protection for chicken embryos,and the survival rate of chicken embryos at 250 mg/mL is 100%.In the test of hemagglutination titer,the hemagglutination titers of the three administration methods at 62.5,125.0 and 250.0 mg/mL were lower than 1/4 of the virus control group,showing antiviral activity(P<0.01),and showing a certain dose-response relationship.Conclusion:The three administration methods of Chinese Herbal Compound Mixture can significantly inhibit the proliferation of influenza A virus A/PR/8/34(H1N1)in chicken embryos,and have a good protective effect on chicken embryos.
