二十五味鬼臼丸对围绝经期综合征大鼠模型血清激素及子宫组织雌激素受体水平的影响

摘　　要：目的研究藏药二十五味鬼臼丸对围绝经期综合征(MPS)大鼠模型血清激素及其子宫组织雌激素受体水平的影响。方法将雌性大鼠随机分为假手术组(Sham)、模型组(0VX)、雌二醇组(OVX+E_(2))、二十五味鬼臼丸组(OVX+GJ)。术后2周时灌胃(假手术组和模型组予等体积生理盐水,雌二醇组予戊酸雌二醇0.1mg/kg,二十五味鬼臼丸组予二十五味鬼臼丸0.36g/kg),隔日给药,连续12周。用ELISA法检测促卵泡生成素(FSH)、黄体生成素(LH)和雌二醇(E_(2))的水平;用RT-PCR及免疫组化法分别检测大鼠子宫组织中雌激素受体α(ERα)、雌激素受体β(ERβ)mRNA和蛋白的表达水平。结果与Sham组比较,0VX组E2表达水平下降(P<0.05),FSH、LH表达水平增高(P<0.05),子宫ERα、ERβmRNA和蛋白表达水平降低(P<0.05);用雌二醇和二十五味鬼臼丸干预后,与0VX组比较,OVX+E_(2)组及OVX+GJ组E_(2)水平增高(P<0.05),FSH、LH水平降低(P<0.05),OVX+E_(2)组子宫ERα、ERβmRNA和蛋白表达水平增高(P<0.05),0VX+GJ组ERαmRNA和蛋白表达水平无差异(P>0.05),ERβmRNA及蛋白表达增高(P<0.05)。结论藏药二十五味鬼臼丸对MPS大鼠模型血清激素及其子宫组织雌激素受体水平存在影响(选择性上调ERβ的表达,对ERα无明显影响,具有“选择性雌激素受体调节剂”作用),这种影响可能是二十五味鬼臼丸有效改善MPS的分子机制之一。Objective To study the effects of Tibetan medicine Guijiu Pill of 25 ingredients on the levels of ser­um hormones and estrogen receptors in uterine tissue with perimenopausal syndrome(PMS)of rat model.Methods Fe­male rats were randomly divided into sham operation group(Sham),OVXgroup(OVX),estradiol group(OVX+E_(2)),and Guijiu Pill of 25 ingredients group(OVX+GJ).The drug was given by gavage 2 weeks after the operation(Sham operation group and OVX group were given equal volume of normal saline,estradiol group was given estradiol valerate 0.1mg/kg,and Guijiu Pill of 25 ingredients group was given Guijiu Pill of 25 ingredients 0.36g/kg)every other day for 12 weeks in a row.ELISA was used to detect the levels of follicle-stimulating hormone(FSH),luteinizing hormone(LH)and estradiol(E_(2));RT-PCR and immunohistochemical methods were used to detect estrogen receptorα(ERα),estrogen receptorβ(ERβ)mRNA and the levels of protein expression.Results Compared with Sham group,the expression of E_(2)in OVX group decreased(P<0.05)and the expression levels of FSH and LH increased(P<0.05).The levels of ERα and ERβ mRNA and protein in the uterus decreased(P<0.05);after the intervention with estradiol and GuijiuPill of 25 ingredients,the E_(2)level of OVX+E_(2)group and the OVX+GJ group increased(P<0.05)compared with OVX group;the levels of FSH and LH decreased(P<0.05);ERα,ERβmRNA in the uterus and protein ex­pression levels increased in OVX+E_(2)group(P<0.05).ERαmRNA and the levels of protein expression in OVX+GJ group were not different(P>0.05).The levels of ERβmRNA and protein expression increased(P<0.05).Conclu­sion The Tibetan medicine Guijiu Pill of 25 ingredients has an effect on the levels of serum hormones and estrogen receptor in the uterine tissue of MPS rat models selectively up-regulating the expression of ERβhas no significant effect on ERα,which had the effect of“selective modulator estrogen receptor”.This effect may be one of the molec­ular mechanisms by which Guijiu Pill of 25 ingredients can im
