阳明病(阳明腑实证)欲解时TNF-α的变化及大承气汤干预作用的实验研究

摘　　要：目的探讨肿瘤坏死因子(TNF-α)在阳明病(阳明腑实证)欲解时及大承气汤干预后变化的研究。方法健康未孕SD大鼠290只,随机分为正常组(n=90)、模型组(n=100)、大承气汤组(n=100)。正常组每天给予等量蒸馏水,模型组采用给实验大鼠灌中药热药、自身粪便混悬液及脂多糖的方法造成大鼠阳明病(阳明腑实证)的模型,大承气汤组在造模成功后分别于卯时(5:00-7:00)、酉时(17:00-19:00)给予大承气汤干预,连续观察5天。造模结束与药物干预后,每天于卯时、酉时从正常组中抽取9只大鼠,模型组和大承气汤组中抽取10只大鼠,断头处死后获取组织匀浆,观察肿瘤坏死因子(TNF-α)含量变化。结果正常组TNF-α表达低于模型组(P<0.01),模型组TNF-α表达呈现节律性,大承气汤组欲解时TNF-α表达有短暂升高后逐渐降低,低于非欲解时组(P<0.05),最后趋于稳定。结论阳明病(阳明腑实证)模型组大鼠TNF-α表达呈现时间节律性,经大承气汤干预后,大承气汤组TNF-α表达逐渐下降,尤以酉时明显。Objective To study the changes of tumor necrosis factor(TNF-α) in Yangming disease(Yangming fuviscera excess syndrome) and after the intervention of Dachengqi Decoction.Methods In total,290 healthy nonpregnant SD rats were randomly divided into normal group(n=90),model group(n=100),and Dachengqi group(n=100).The normal group was given the same amount of distilled water every day.The model group was fed with hot traditional Chinese medicine(TCM) decoction,fecal suspensions,and lipopolysaccharide to establish the model of Yangming disease.After the successful establishment of the model,the rats in the Dachengqi group were given Dachengqi Decoction for intervention at 5:00-7:00 and 17:00-19:00 and observed for 5 consecutive days.After the completion of model making and drug intervention,9 rats were extracted from the normal group,10 rats were extracted from the model group and Dachengqi group every day,tissue homogenates were obtained after decapitation,and the content changes of tumor necrosis factor(TNF-α) were observed.Results The expression of TNF-α in the normal group was lower than that in the model group(P <0.01),and the expression of TNF-α in the model group presented a rhythmical pattern.The expression of TNF-α in the Dachengqi group briefly increased and then gradually decreased,which was lower than that in the non-desiring group(P <0.05),and finally tended to be stable.Conclusion The expression of TNF-α in the model group of Yangming disease(Yangming fu-viscera excess syndrome) was rhythmical,and after the intervention of Dachengqi Decoction,the expression of TNF-α in the Dachengqi group decreased gradually,especially in the period of the day from 17:00 to 19:00.
