Effects of Fried Semen Ziziphi Spinosae on Information Coding of Basolateral Amygdala Neurons in Rats with State Anxiety

摘  要:目的:探讨炒酸枣仁对状态性焦虑大鼠的行为学及基底外侧杏仁核(Basolateral amygdala, BLA)神经元信息编码的影响。方法:SD雄性大鼠随机分为模型对照组与炒酸枣仁组,灌胃给予炒酸枣仁组大鼠炒酸枣仁水煎液(17.5 g/kg),模型对照组灌服相同体积的蒸馏水,给药0.5 h后采用旷场实验方法及在体多通道神经同步记录技术,监测两组大鼠的行为学及BLA区神经元动作电位放电情况。结果:与模型对照组相比,炒酸枣仁组大鼠在旷场中的跨格次数、站立次数及总运动时间无明显变化(P>0.05),中央格运动时间、进入中央格次数及中央格/总运动时间(%)明显增加(P<0.01)。与自身的基础放电频率比较,两组大鼠在旷场中的BLA区Ⅰ类神经元(NeuronⅠ)、Ⅱ类神经元(NeuronⅡ)动作电位放电频率均有所增加。与模型对照组相比,炒酸枣仁组大鼠NeuronⅠ动作电位放电频率增加值明显减少(P<0.05),NeuronⅡ动作电位放电频率增加值无明显变化(P>0.05)。同时,大鼠在旷场中的中央格运动/总运动时间(%)与BLA区NeuronⅠ、NeuronⅡ动作电位放电频率增加值均呈明显的负相关(P<0.05或P<0.01)。结论:炒酸枣仁能明显降低大鼠在旷场中的焦虑样行为,具有一定的拮抗状态性焦虑作用,其作用机制与抑制BLA区焦虑相关神经元动作电位放电频率、改变BLA区神经元信息编码有关。Objective:To investigate the effects of fried Semen Ziziphi Spinosae on behavior and information coding of basolateral amygdala(BLA)neurons in rats with state anxiety.Methods:Male SD rats were randomly divided into the model control group and the fried Semen Ziziphi Spinosae group.Rats in the fried Semen Ziziphi Spinosae group were given decoction of fried Semen Ziziphi Spinosae by gavage(17.5 g/kg).Rats in the model control group were given the same amount of volume of distilled water.After 0.5 h of administration,open field experiment and in vivo multi-channel neural synchronous recording technology were used to monitor the behavior and spike firing rate of BLA region in the two groups.Results:Compared to those in the model control group,the cross grid times,standing times and total exercise time had no significant changes in the fried Semen Ziziphi Spinosae group(P>0.05);however,the central grid movement time,the number of entering the central grid and the central grid/total exercise time(%)were significantly increased(P<0.01).Compared to the basic firing rate,the spike firing rate of type I and type II neurons were increased in BLA region of the open field in the two groups.Compared to that in the model control group,the increase value of spike firing rate of type I neurons in BLA region was significantly decreased in the fried Semen Ziziphi Spinosae group(P<0.05);whereas the increase value of spike firing rate of typeⅡneurons had no significant change(P>0.05).Besides,there was a significant negative correlation between the time of central grid movement/total motor time(%)and the increase of spike firing rate of type I and type II neurons in BLA region(P<0.05,P<0.01).Conclusion:Fried Semen Ziziphi Spinosae could significantly reduce the anxiety-like behavior of rats in open field,and had a certain improving effect on state anxiety.The mechanism is related to the inhibition of spike firing rate of anxiety related neurons and the change of neuronal information coding in BLA region.
