摘 要:目的:观察穴位埋线疗法对氯化锂-匹罗卡品致痫大鼠认知行为学、镜下海马CAl区神经元丢失程度半定量分析及脑源性神经营养因子(BDNF)蛋白表达的影响。方法:80只健康SD雄鼠随机分为空白组、模型组、西药组及埋线组,每组20只。癫痫模型建立后分别进行相应的干预共28 d,选择干预前及末次干预2个时间点,分别对各组大鼠进行拒俘反应实验;到达预定存活时间时,处死各组大鼠取海马,尼氏染色法半定量分析各组海马CAl区的神经细胞丢失程度,免疫组化法检测各组海马CA1区BDNF蛋白表达的情况。结果:西药组及埋线组大鼠攻击行为和逃避行为明显减少(P<0.05);西药组、埋线组神经元丢失程度有所减轻(P<0.05);西药及穴位埋线干预后海马中BDNF表达均较模型组增高(P<0.05);各指标西药组与埋线组比较,差异无统计学意义(P>0.05)。结论:癫痫可造成大鼠认知障碍,穴位埋线可以通过间接提高BDNF蛋白表达以减缓海马神经细胞丢失程度,具有明显的改善癫痫后认知障碍的作用,疗效与口服抗癫痫药左乙拉西坦相当。Objective:To observe the effects of acupoint embedding on cognitive behavior,hippocampal neuron loss and expression of brain-derived neurotrophic factor(BDNF)in epilepsy rats induced by Lithium Chloride-Pilocarpine.Methods:80 male SD rats were randomly divided into the blank group,the model group,the western medication group and the acupoint embedding group,with 20 rats in each group.The epilepsy model was established by intraperitoneal injection of Lithium Chloride-Pilocarpine.The corresponding interventions were performed for 28 days after successful modeling.The captive reaction experiments were performed before and after the intervention in each group.The neuron loss of the hippocampal CA1 region was analyzed by Nissl staining.The protein expression of BDNF in the hippocampal CA1 region was detected by immunohistochemical method.Results:The aggressive behavior and the escape behavior of the model group were increased than those of the blank group(P<0.01);after the interventions of western medication and acupoint embedding,the aggressive behavior and the escape behavior were significantly reduced(P<0.05).The neuron loss was significant in the model group compared to that in the blank group(P<0.01);which was relieved after the interventions of western medication and acupoint embedding(P<0.05).The protein expression of BDNF was significantly reduced in the model group compared to that in the blank group;which was significantly increased after the interventions of western medication and acupoint embedding(P<0.05).There were no statistical differences in the aggressive behavior and the escape behavior,the neuron loss and the protein expression of BDNF between the western medication group and the acupoint embedding group(P>0.05).Conclusion:Epilepsy can cause cognitive impairment in rats.Acupoint embedding can reduce the loss of hippocampal neuron by increasing the protein expression of BDNF indirectly.Acupoint embedding has an obvious effect on improving cognitive impairment after epilepsy,and the efficacy is equal
