摘  要:目的研究携Pik3cb shRNA的磁性纳米微泡在磁场和超声双重作用下,对受损伤血管壁中平滑肌细胞的影响。方法构建携Pik3cb shRNA的磁性纳米微泡,检测其粒径和Zeta电位。建立大鼠颈总动脉球囊损伤模型,随机分为磁性纳米微泡组和转染对照组。2周后麻醉处死取损伤处血管进行HE染色和免疫组化ki67染色观察受损伤血管内膜增生的程度和平滑肌细胞的增殖情况。结果制得的携shRNA的磁性纳米微泡的平均粒径为(1.75±0.32)μm,Zeta电位为(10±0.79)mV。磁性纳米微泡组的内膜增厚程度和平滑肌细胞的增殖程度较转染对照组减轻.结论成功构建了携Pik3cb shRNA的磁性纳米微泡,在磁场和超声双重作用下,可以实现其对平滑肌细胞的靶向高效转染,发挥抑制血管再狭窄的作用。Objective To examine the transfection efficacy of superparamagnetic iron oxide(SPIO)nanoparticles carrying with Pik3cb shRNA in vascular smooth muscle cells(VSMCs)of the injured artery under the dual function of magnetic field and ultrasound.Methods SPIO nanoparticles carrying with Pik3cb shRNA were prepared and their particle size and Zeta potential were measured.20 balloon-injured rats were randomly divided into 2 groups:The treat-ment group received SPIO nanoparticles carrying Pik3cb shRNA,the control group received saline.Rats were sacri-ficed and tissue were harvested after two weeks,HE staining and immunohistochemistry analysis were used to observe the intimal hyperplasia and the proliferation of VSMCs.Results The average particle size of the SPIO nanoparticles carrying with shRNA was(1.75±0.32)μm,and the Zeta potential was(10±0.79)mV.Compared with the control group,the intimal thickening in the treatment group was decreased and the proliferation of VSMCs was extenuated.Conclusions SPIO nanoparticles carrying with Pik3cb shRNA are successfully prepared and enhanced the transfection efficiently under the dual function of magnetic field and ultrasound.These novel nanoparticles have potential therapeu-tic value for the treatment for restenosis.
