摘 要:目的:建立大鼠铜绿假单胞菌(PA)肺部感染模型,并对模型进行初步评价。方法:从大鼠切开的气管内注入PA琼脂糖包裹体(实验组)或等量无菌琼脂糖包裹体(对照组)。术后第1、第3、第5天检测两组肺部细菌含量,苏木精-伊红(HE)染色观察肺组织病理学改变,酶联免疫吸附试验(ELISA)法检测血清和肺组织中肿瘤坏死因子(TNF)-α和白细胞介素(IL)-17A含量。结果:实验组肺组织中检出PA,肺部有明显炎症,第1天肺组织以多形核白细胞(PMN)浸润为主,第5天PMN减少,单核巨噬细胞、淋巴细胞等慢性炎症细胞增多;对照组肺组织未见PA,肺部无明显炎症;接种后第1、第3、第5天,实验组大鼠TNF-α、IL-17A含量高于对照组(P<0.05),实验组接种后第5天TNF-α、IL-17A含量均高于接种后第1天(P<0.01)。结论:气管切开注入PA琼脂糖包裹体,可成功建立大鼠PA肺部感染模型。该模型操作简单,重复性好,可用于进行PA急慢性肺部感染的各种研究。Objective:To establish and preliminarily validate an animal model of pulmonary Pseudomonas aeruginosa(PA)infection by tracheotomy.Methods:The PA agarose inclusion(experiment group)or the same volume of sterile agarose inclusion(control group)was injected into the trachea of the rats through tracheotomy.The content of bacteria in the lungs on the 1st,3rd and 5th day after the operation was detected.The pathological changes of lung were observed by hematoxylin-eosin(HE)staining.The levels of tumor necrosis factor(TNF)-αand interleukin(IL)-17A in serum and lung tissues were determined by enzyme-linked immunosorbent assay(ELISA).Results:PA was detected in the lung tissue of the experimental group,and there was obvious lung inflammation.On day 1,the lung tissue was mainly infiltrated by PMN.On day 5,PMN decreased and chronic inflammatory cells such as mononuclear macrophages and lymphocytes increased.No PA was detected in the lung tissue in the control group,and no obvious inflammations were observed in pulmonary pathology.On the 1st,3rd and 5th day after inoculation,the levelsofTNF-αand IL-17A of the experimental group were significantly higher than that of the control group(P<0.05).In the experimental group,the levels of TNF-αand IL-17A on day 5 were higher compared with day 1(P<0.01).Conclusion:The rat model of pulmonary PA infection can be successfully established by injecting PA agarose inclusion through tracheotomy.The model is easy to operate and has good repeatability,which is applicable for the study of PA acute and chronic pulmonary infection.
