连夏配方颗粒对心肌梗死大鼠室颤阈值和连接蛋白43重构的影响

摘　　要：目的:探讨连夏配方颗粒对心肌梗死大鼠室颤阈值和连接蛋白43(Cx43)重构的影响。方法:通过结扎大鼠左冠状动脉前降支建立心肌梗死模型,将造模成功的大鼠随机分为模型组、连夏配方颗粒高、中、低剂量组和美托洛尔组,另设假手术组。干预30 d后采用在体程控电刺激检测大鼠室颤阈值;RT-PCR法检测左心室心梗边缘区P38mRNA、Cx43mRNA的表达;免疫组织化学染色检测Cx43的密度和分布;酶联免疫吸附试验(ELISA)检测血清肿瘤坏死因子-α(TNF-α)的含量。结果:与假手术组比较,模型组大鼠室颤阈值显著降低,Cx43mRNA表达下降,分布紊乱,血清TNF-α的含量和心肌P38mRNA表达显著增加(P<0.01);与模型组比较,连夏配方颗粒高、中剂量组大鼠室颤阈值提高(P<0.01,P<0.05),Cx43mRNA表达增加(P<0.05),Cx43分布改善,血清TNF-α的含量和心肌P38mRNA的表达显著降低(P<0.01)。结论:连夏配方颗粒可提高心梗大鼠室颤阈值,其机制可能与减轻炎症反应,改善连接蛋白43重构有关。Objective:To investigate the effect of LianXia Formula Granule on ventricular fibrillation threshold and connexin 43(Cx43)remodeling in myocardial infarction rats.Methods:The model of myocardial infarction was established by ligating the left anterior descending coronary artery of the rat,and the successfully modeled rats were randomly divided into a model group,a Lianxia formula granule high,medium,and low dose group,and a metoprolol group,and a sham operation group.After 30 days of intervention,the ventricular fibrillation threshold was detected by in vivo programmed electrical stimulation;RT-PCR method was used to detect the expression of P38mRNA and Cx43mRNA in the marginal zone of left ventricular myocardial infarction;the density and distribution of Cx43 were detected by immunohistochemical staining;the content of serum TNF was detected by ELISA method.Results:Compared with the sham operation group,the ventricular fibrillation threshold of rats in the model group was significantly reduced,the expression of Cx43mRNA was decreased,the distribution was disordered,the content of serum TNF-αand the expression of myocardial P38mRNA were significantly increased(P<0.01).Compared with the model group,the ventricular fibrillation threshold of rats in the high and medium dose groups of Lianxia formula particles increased(P<0.01,P<0.05),the expression of Cx43mRNA increased(P<0.05),the distribution of Cx43 was improved,and the level of serum TNF-αand the expression of P38mRNA in myocardium was significantly reduced(P<0.01).Conclusion:LianXia Formula Granule can raise ventricular fibrillation threshold,and its mechanism is related to reducing inflammation and improving connexin 43 remodeling.
