摘  要:为研究增强丝棉木组培苗的抗逆性方法,以丝棉木1 a生休眠枝条带1~2个腋芽的茎段为外植体,经外植体表面消毒、腋芽萌发诱导并利用抗生素进行组培苗脱菌处理,研究不同抗生素种类、浓度及不同继代培养次数对丝棉木组培苗抑菌效果及其生长的影响,探索适合丝棉木组培脱菌的最佳方法。结果表明:1/2 MS+50mg/L制霉菌素+50mg/L硫酸链霉素+250mg/L羧苄青霉素钠能够有效抑制丝棉木外植体真菌与细菌污染,丝棉木外植体在该培养基上每隔20 d继代培养1次,3~4次继代培养后,并经生根培养后可获得生长势强的脱菌苗。In order to study the methods of enhancing the stress resistance of tissue culture seedlings of Euonymus maackii,the stem segments with 1-2 axillary buds of a-year-old dormant branches were used as explants.After surface disinfection of explants,axillary bud germination induction and antibiotic treatment,the effects of different antibiotic types,concentrations and subculture times on the antibacterial effect and growth of tissue culture seedlings of E.maackii were studied,and the best method suitable for tissue culture of E.maackii was explored.The results showed that the 1/2MS medium containing 50 mg/L nystatin,50 mg/L streptomycin sulfate and 250 mg/L carbenicillin disodium could effectively inhibit the explants of E.maackii in vivo fungus and bacterial contamination,and the explants of E.maackii were subcultured once every 20 days,and after 3 to 4 times of subculture and rooting culture,the strong growth potential of the bacteria-free seedlings could be obtained.
