摘 要:目的研究黄芩苷在体外对牙龈卟啉单胞菌(Porphyromonas gingivalis, P.g)、具核梭杆菌(Fusobacterium nucleatum, F.n)及伴放线聚集杆菌(Aggregatibacter actinomycetemcomitans, A.a)的抑菌作用,通过大鼠实验性牙周炎模型评价其抑制破骨细胞生成及抗炎效果。方法采用倍比稀释法,测定黄芩苷对P.g、F.n、A.a的最低抑菌浓度(MIC)及最低杀菌浓度(MBC)。将42只SD大鼠随机分为2%盐酸米诺环素组(IM组)、0.01%黄芩苷组(BL组)、0.1%黄芩苷组(BM组)、1%黄芩苷组(BH组)、甘油溶剂组(Tri组),牙周炎模型组(EP组)、正常组(NG组)。测定各组体质量、探诊深度(PD)、龈沟出血指数(SBI),进行HE染色,用抗酒石酸酸性磷酸酶(TRAP)染色检测破骨细胞数量及酶联免疫吸附法(ELISA)测定血清中环氧合酶2(COX-2)、基质金属蛋白酶9(MMP-9)水平。结果黄芩苷对P.g、F.n、A.a的MIC分别为1.0、2.0、2.0 g/L,MBC分别为2.0、8.0、4.0 g/L。各组PD、SBI均在早期呈现快速上升的趋势,BM、BH、IM组上下浮动的趋势大致相同。HE染色中,正常牙龈组织内无炎症细胞浸润,EP组与Tri组中有大量的炎症细胞浸润及上皮的破坏。TRAP染色结果显示牙槽骨破骨细胞的数量随着药物浓度的增加而减少。与NG组相比,EP组及Tri组中血清COX-2、MMP-9水平在中浓度升高,差异有统计学意义(P<0.05)。与EP组及Tri组相比,BL、BM、BH、IM组中血清COX-2、MMP-9水平降低(P<0.05),差异有统计学意义。结论黄芩苷对P.g、F.n及A.a均有抑制作用,可有效减少牙周组织中减少破骨细胞数量,降低实验性牙周炎大鼠血清中的COX-2、MMP-9水平。Objective To study the bacteriostatic effect of baicalin on Porphyromonas gingivalis(P.g),Fusobacterium nucleatum(F.n)and Aggregatibacter actinomycetemcomitans(A.a)in vitro,and to evaluate the inhibition of osteoclast formation and anti-inflammatory effect in rats with experimental periodontitis.Methods The minimum inhibitory concentration(MIC)and minimum bactericidal concentration(MBC)of baicalin to P.g,F.n and A.a were determined bymultiple dilution;a total of 42 SD rats were randomly divided into 2%minocycline group(IM),0.01%baicalin group(BL),0.1%baicalin group(BM),1%baicalin group(BH),1%baicalin group(BH),glycerol solvent group(Tri),peri-odontitis model group(EP)and normal group(NG).Body weight,probing depth(PD)and sulcus bleeding index(SBI)were measured.The number of osteoclasts was detected by tartrate resistant acid phosphatase(TRAP)staining and HEstaining,and the levels of cyclooxygenase-2(COX-2)and matrix metalloproteinase-9(MMP-9)in serum were mea-sured by enzyme-linked immunosorbent assay(ELISA).Results The MIC of baicalin to P.g,F.n and A.a were 1.0 g,2.0 g,2.0 g,2.0 g,8.0 g and 4.0 g respectively.PD and SBI in all groups showed a rapid upward trend in the early stage,and the upward and downward trends in BM,BH and IM groups were roughly the same.In HE staining,there was noinflammatory cell infiltration in normal gingival tissue,but there were a large number of inflammatory cell infiltrationand epithelial destruction in EP and Tri groups.TRAP staining showed that the number of osteoclasts in alveolar bonedecreased dose-dependently.Compared with the normal group,the serum levels of COX-2 and MMP-9 in the EP andTri groups were significantly increased(P<0.05).Compared with the EP and Tri groups,the serum levels of COX-2 and MMP-9 in BL,BM,BH and IM groups were significantly lower(P<0.05).Conclusions Baicalin can inhibit P.g,F.n and A.a.It can effectively reduce the number of osteoclasts in periodontal tissue and reduce the levels of COX-2 and MMP-9 in the serum of experimental periodontitis rats.
