摘  要:本研究以甘孜藏族自治州野生手参为试验材料,研究不同外植体类型(茎尖、根尖、侧芽)和植物生长调节剂(2,4-D、6-BA、NAA)对手参愈伤组织诱导的影响,并检测愈伤组织中药用化学成分含量。结果表明,茎尖的愈伤组织诱导率高于根尖和侧芽,在MS+NAA 0.5 mg/L培养基中手参愈伤组织诱导率最高(59.5%),且单独添加生长素时愈伤组织诱导率高于组合添加同一浓度的生长素和细胞分裂素;根尖的褐化率最高(100.0%),茎尖的褐化率最低(38.5%);附加NAA 0.5 mg/L的培养基诱导的愈伤组织天麻素和dactylorhin A含量均高于附加2,4-D 0.5 mg/L的培养基诱导的愈伤组织。本研究结果可为进一步提高手参的品质,增强我国药用植物产业的竞争力,为后续商业化应用打下坚实的基础。In this study,using the wild Gymnadenia conopsea in GarzêTibetan Autonomous Prefecture as the experimental material,the effects of different types of explants(shoot apex,root apex,side bud)and plant growth regulators(2,4-D,6-BA,NAA)on callus induction of Gymnadenia conopsea were studied,and the content of medicinal chemical components in the callus was detected.The results showed that the callus induction rate of shoot apex was the higher than that of root apex and side bud,the callus induction rate of Gymnadenia conopsea was the highest(59.5%)in MS+NAA 0.5 mg/L medium,and the callus induction rate was higher when auxin was added alone than that of the same concentration of auxin combined with cytokinin.The browning rate of root apex was the highest(100.0%),and that of shoot apex was the lowest(38.5%);the contents of gastrodin and dactylorhin A in callus induced by medium added NAA 0.5 mg/L were higher than those in callus induced by medium added 2,4-D 0.5 mg/L.The results of this study could further improve the quality of Gymnadenia conopsea,enhance the competitiveness of China′s medicinal plant industry,and lay a solid foundation for subsequent commercial application.
