摘 要:目的研究补肾安胎合剂对肾虚流产小鼠EPCs动员、归巢的影响。方法对100只ICR小鼠作进行性骨髓移植+肾虚流产造模,分为空白组、模型组及补肾安胎合剂高、中、低剂量组。灌胃给药4周后,剖视小鼠子宫检测胚胎丢失率,流式细胞学检测骨髓、外周血、蜕膜组织内皮祖细胞(EPCs)数,ELISA法检测外周血基质细胞衍生因子-1α(SDF-1α)水平和蜕膜组织趋化性细胞因子受体-4(CXCR-4)表达。结果骨髓移植模型造模成功率为53.2%。与空白组比较,模型组、中药低剂量组胚胎丢失率升高(P<0.01);与模型组比较,中药高、中剂量组丢失率降低(P<0.01)。与空白组比较,各流产组骨髓、外周血、蜕膜组织中EPCs数目降低(P<0.05,P<0.01);与模型组比较,中药高剂量组EPCs数目升高(P<0.01)。在外周血中,与空白组比较,模型组及中药低、中剂量组SDF-1α水平降低(P<0.05,P<0.01);与模型组比较,各中药组SDF-1α水平升高(P<0.01)。在蜕膜组织中,与空白组比较,各流产组CXCR-4水平降低(P<0.01);与模型组比较,中药高、中剂量组蜕膜组织中CXCR-4水平升高(P<0.01)。结论补肾安胎合剂可能通过提高外周血SDF-1α水平及蜕膜组织CXCR-4表达来促进EPCs动员与归巢,改善母胎界面血管重铸,从而降低胚胎丢失率。AIM To investigate the effects of Bushen Antai Mixture on EPCs mobilization and homing in mice with abortion due to Kidney Deficiency.METHODS One hundred ICR mice were conducted bone marrow transplantation and Kidney Asthenia abortion models were constructed.The bone marrow transplantation mice were assigned into the blank group,and the group for development of Kidney Asthenia abortion models and subsequently assigned into model group,Bushen Antai Mixture groups(high,medium and low doses)for,4-week corresponding gavage,after which the following items were performed:the detection of the rate of mouse embryo loss by hysterotomy;the determination of EPCs count in bone marrow,peripheral blood and decidual tissues by flow cytometry;the identification of the levels of SDF-1αin peripheral blood and CXCR-4 in decidual tissues by ELISA.RESULTS The success rate of bone marrow transplantation was 53.2%.The model group and the low-dose Bushen Antai Mixture group displayed higher embryo loss rate than the blank group(P<0.01).The high-dose and medium-dose Bushen Antai Mixture groups had lower embryo loss rate than the model group(P<0.01).The abortion groups shared less EPCs count in the bone marrow,peripheral blood and decidual tissues than the blank group(P<0.05,P<0.01).The high-dose Bushen Antai Mixture group had more EPCs count than the model group(P<0.01).Compared with the blank group,the model group,the low-dose and medium-dose Bushen Antai Mixture groups demonstrated less SDF-1αlevel in the peripheral blood(P<0.05,P<0.01),but more SDF-1αlevel was observed in every Bushen Antai Mixture group than that in the model group(P<0.01).CXCR-4 expression in every abortion group was lower than that in the blank group(P<0.01),and CXCR-4 expressions in the decidual tissues in the high-dose and medium-dose Bushen Antai Mixture groups were higher than that in the model group(P<0.01).CONCLUSION Bushen Antai Mixture may promote the mobilization and homing of EPCs by increasing the level of SDF-1αin peripheral blood and the expression
