摘 要:为了提高柚皮苷的稳定性及其生理活性,利用微波辅助非水相酶催化反应完成柚皮苷的乙酰化合成。考察并优化了影响柚皮苷微波酶促乙酰化反应的主要因素,以及其对HepG2细胞的抗增殖影响。结果表明,微波功率为400W、微波温度为70℃、水活度为0.80时柚皮苷酶促乙酰化反应的最高酶活为(612±10)μmol/(h·g),比传统方法提高4倍左右,且在2h内的转化率为90.42%±2.43%,经核磁波谱仪鉴定反应产物为6′-乙酰化柚皮苷。虽与母体柚皮苷相比,乙酰化柚皮苷的体外抗氧化活性略微降低,但对HepG2细胞具有较强的抗增殖能力。表明,柚皮苷非水相酶促乙酰化反应具有生物学意义。Acetylated naringin was synthesized by microwave-assisted non-aqueous enzymatic reaction to improve the stability and physiological bioactivity.The effects of various parameters on the enzyme activity and anti-proliferation effect on HepG2 were investigated and optimized.The results showed that the enzyme activity reached(612±10)μmol(/h·g)under the optimum conditions of microwave power 400 W,microwave temperature 70℃and water activity 0.80,which was about 4 times higher compared to traditional reaction.The maximum conversion was(90.42±2.43)%after reaction 2 h.The product was identified as 6’-acetylnaringin by nuclear magnetic resonance(NMR),the mass fraction was larger than 99%.Although the antioxidant activity of acetylated naringin in vitro was slightly lower than that of maternal naringin,it had a strong antiproliferative activity against HepG2 cells.That meant that the non-aqueous enzymatic acetylation of naringin has biological significance.
