摘 要:目的分析2017年10月—2019年9月新疆B型流感病毒NA基因,为流感NAI抑制剂的使用提供依据。方法选取2017年10月—2019年9月分离的9株B/Yamagata系毒株和13株B/Victoria系毒株,采用逆转录—聚合酶链反应(RT-PCR)扩增病毒NA基因后测定核苷酸序列,用MEGA 5.0,CExpress和BioEdit生物软件分析测序结果、基因特性并建树。结果13株Victoria系B型毒株与疫苗株B/Brisbane/60/2008核苷酸同源性为97.9%~100.0%,核苷酸进化树显示,13株Victoria系B型毒株与疫苗株为同一分支,且远离国外B型流感耐药株B/North Carolina/03/2011 I21V和B/North Carolina/11/2010 I21V;9株Yamagata系B型毒株与疫苗B/Phuket/3073/2013株核苷酸同源性为96.4%~99.9%;9株Yamagata系B型毒株与疫苗株为同一分支,且远离国内耐药株B/Beijing-Xicheng/11496/2011 D197N和国外B型流感耐药株B/Ontario/006876/2011 H273Y;所有毒株的NA蛋白酶催化位点和辅助位点,均未发现耐药位点的替换。结论2017—2019年新疆B型流感病毒NA基因未发生耐药位点突变,与对应疫苗株的核苷酸同源性较高,仍对NAI抑制剂敏感;应继续加强流感监测。Objective To analyze the NA Gene of influenza B virus in Xinjiang from October 2017 to September 2019,and provide basis for application of NAI inhibitors of influenza.Methods Nine strains of B/Yamagata virus and 13 strains of B/Victoria virus isolated from October 2017 to September 2019 were selected for nucleotide sequencing after NA gene amplified by RT-PCR.The MEGA 5.0,CExpress and BioEdit softwares were taken for analyzing the nucleotide sequence,gene characteristics and establishing the gene tree.Results The nucleotide homologies between the 13 strains of B/Victoria virus and the vaccine strain of B/Brisbane/60/2008 were from 97.9%to 100.0%.The nucleotide phylogenetic tree showed both the 13 strains of B/Victoria virus and the vaccine strain belonging to the same branch,and were far away from the drug resistant strain of foreign influenza B(B/North Carolina/03/2011 I21 V and B/North Carolina/11/2010 I21 V).The nucleotide homologies between the 9 strains of B/Yamagata virus and the vaccine strain of B/Phuket/3073/2013 were from 96.4%to 99.9%.Both the 9 strains of B/Yamagata virus and the vaccine strain belonged to the same branch,and were far away from the drug-resistant strains of domestic B/Beijing-Xicheng/11496/2011 D197 N and foreign B/Ontario/006876/2011 H273 Y.The catalytic loci and the subsidiary loci of NA protease in all strains were not substituted by the drug resistance loci.Conclusions There is no mutation of drug resistance loci in NA gene of influenza B virus in Xinjiang from 2017 to 2019,having high nucleotide homologies with the corresponding vaccine strain,and the virus are still sensitive to the NAI inhibitors.The influenza surveillance should be continuously strengthened.
