摘  要:为了研究抗氧化剂绿原酸(CGA)、咖啡酸苯乙酯(CAPE)及冷冻保护剂聚乙烯吡咯烷酮(PVP)联合添加对猪冷冻精液品质的影响,试验采用手握法对3头新吉林黑猪进行精液采集,将品质最佳的精液平均分为7组,即对照组和试验Ⅰ、Ⅱ、Ⅲ、Ⅳ、Ⅴ、Ⅵ组,其中对照组不添加抗氧化剂及冷冻保护剂,试验Ⅰ组添加100μmol/L CGA,试验Ⅱ组添加40μmol/L CAPE,试验Ⅲ组添加0.5 g/L PVP,试验Ⅳ组添加100μmol/L CGA和0.5 g/L PVP,试验Ⅴ组添加40μmol/L CAPE和0.5 g/L PVP,试验Ⅵ组添加100μmol/L CGA、40μmol/L CAPE和0.5 g/L PVP,进行液氮冷冻保存,30 d后解冻精液,测定精子活力、线粒体活率、DNA完整率、质膜完整率、顶体完整率、过氧化氢酶(CAT)活性、超氧化物歧化酶(SOD)活性、谷胱甘肽过氧化物酶(GSH-Px)活性、活性氧(ROS)水平及丙二醛(MDA)含量。结果表明:与对照组相比,添加100μmol/L CGA、40μmol/L CAPE和0.5 g/L PVP的冷冻精液的精子活力、质膜完整率、顶体完整率、线粒体活率、CAT活性、GSH-Px活性均极显著提高(P<0.01);DNA完整率、SOD活性均显著提高(P<0.05);ROS水平极显著降低(P<0.01);MDA含量显著降低(P<0.05)。说明在猪精液冷冻基础液中同时添加100μmol/L CGA、40μmol/L CAPE和0.5 g/L PVP可提高新吉林黑猪冷冻精液品质。To investigate the effects of the combination of antioxidant chlorogenic acid(CGA), phenethyl caffeate(CAPE) and cryoprotectant polyvinylpyrrolidone(PVP) on the quality of frozen semen in pigs, semen from 3 Xin Jilin Black pigs were collected by hand grip method in this experiment. The semen with the best quality was divided into 7 groups on average, namely control group(no antioxidant and cryoprotectant), experimental group I(add 100 μmol/L CGA), experimental group II(add 40 μmol/L CAPE);experimental group III(0.5 g/L PVP), experimental group IV(add 100 μmol/L CGA and 0.5 g/L PVP), experimental group V(add 40 μmol/L CAPE and 0.5 g/L PVP), experimental group VI(add 100 μmol/L CGA、40 μmol/L CAPE and 0.5 g/L PVP). After 30 d of frozen storage in liquid nitrogen, semen was thawed, and the sperm motility, mitochondrial activity rate, DNA integrity rate, plasma membrane integrity rate, acrosome integrity rate, catalase(CAT) activity, superoxide dismutase(SOD) activity, glutathione peroxidase(GSH-Px) activity, reactive oxygen species(ROS) level, and malondialdehyde(MDA) content of frozen semen were measured. The results showed that the sperm motility, plasma membrane integrity rate, acrosome integrity rate, mitochondrial activity rate, CAT activity and GSH-Px activity of frozen semen in adding 100 μmol/L CGA、40 μmol/L CAPE and 0.5 g/L PVP group were significantly improved compared with the control group(P<0.01);DNA integrity rate and SOD activity were significantly increased(P<0.05);the ROS level decreased significantly(P<0.01);and the MDA content decreased significantly(P<0.05). The results indicated that adding 100 μmol/L CGA、40 μmol/L CAPE and 0.5 g/L PVP to the frozen base solution of pig spermatozoa could improve the frozen semen quality of Xin Jilin Black pig.
