摘 要:为了探讨龙牧801号紫花苜蓿(Medicago sativa L.)种子活力及内生细菌对不同浓度苦参碱水溶液的响应,试验采用浓度为0(CK),50,100,200,300,400,500,600 mg/L的苦参碱浇灌紫花苜蓿种子,测定和计算苜蓿种子发芽势、发芽率、发芽指数、活力指数,以及幼苗的胚芽长、胚根长/胚芽长、鲜重及内生细菌数量。结果表明:龙牧801号种子的发芽率、发芽势、发芽指数、活力指数随苦参碱浓度的增加大体上呈先增加后降低趋势(其中活力指数于100 mg/L时降低),均在浓度为200 mg/L时达到最高,分别高于CK 12.18%、20.07%、36.63%和46.69%,差异显著(P<0.05),当苦参碱浓度增加至600 mg/L时各指标降至最低。苦参碱浓度为200 mg/L幼苗胚芽长高于CK 12.80%,差异显著(P<0.05),但抑制幼苗胚根长(P<0.05)。种子内生细菌数量丰富,当苦参碱浓度为200 mg/L时数量最多,高于CK 30.54%。说明苦参碱对龙牧801号紫花苜蓿种子活力和内生细菌有一定促进作用,适宜浓度苦参碱在激发苜蓿种子活力方面具有较大潜力。In order to investigate the response of seed vigor and endophytic bacteria of alfalfa(Medicago sativa. Longmu No. 801) to the different concentrations of matrine, matrine with concentration of 0(CK), 50, 100, 200, 300, 400, 500 and 600 mg/L was used to irrigated alfalfa seeds, and the germination rate, germination potential, germination index and vigor index of alfalfa seed, embryo length, radicle length, ratio of radicle length to embryo length of seedling were measured, and number of endophytic bacteria was calculated. The results showed that the germination rate, germination potential, germination index and vigor index of Longmu No.801 seeds were all increased first and then decreased with the increase of matrine concentration(except the activity index decreased at 100 mg/L), when the concentration reached to 200 mg/L, the above items reached highest, which were significantly higher than that of CK group 12.18%, 20.07%, 36.63% and 46.69%, respectively(P<0.05), the all items dropped to the lowest at the concentration of 600 mg/L. The seedlings embryo length in the 200 mg/L matrine was increased by 12.80%(P<0.05) compared with that in CK group, but the radicle length was significantly inhibited(P<0.05). The number of endophytic bacteria in seeds was abundant, it reached highest at 200 mg/L matrine treatment, which was higher than CK 30.54%. The results indicated that matrine had certain promoting effect on the seed activity and endophytic bacteria of Longmu No.801 alfalfa, and the appropriate concentration of matrine had great potential in stimulating the alfalfa seed activity.
